目的 采用双样本孟德尔随机化方法研究口腔微生物与阿尔茨海默病(Alzheimer's disease,AD)风险之间的因果关系,解决观察性研究中固有的混杂偏倚问题.方法 分析来自488 285 名欧洲后裔的全基因组关联数据.通过逆方差加权(inverse variance weighting,IVW)、加权中位数、MR-Egger回归、简单众数法和加权众数法分析数据.进行敏感性分析校正和排除验证.结果 IVW方法揭示了链球菌 ASV0006 丰度与 AD 易感性之间的显著因果关系(OR=1.000 157,95%CI=1.000 01 9~1.000 295,P=0.025).结论 孟德尔随机化分析结果显示,口腔链球菌(ASV0006)丰度与AD风险增加存在显著的正向关联.链球菌ASV0006 丰度升高可能是AD的一个潜在可调节风险因素.

Objective To investigate the causal relationship between oral microbiota and Alzheimer′s disease(AD)risk with two-sample Mendelian randomisation and to address the problem of confounding bias inherent in observational studies.Methods We analyzed genome-wide association data from 488 285 individuals of European descent.Analyses were performed by inverse variance weighting(IVW),weighted median,MR-Egger,simple mode and weighted mode methods.Sensitivity analyses were corrected and exclusion validation.Results The IVW approach revealed a significant causal relationship between Streptococcus ASV0006 abundance and AD susceptibility(OR=1.000 157,95%CI=1.000 01 9-1.000295,P=0.025).Conclusion Men-delian randomization analysis results showed a significant positive association between the abundance of oral Streptococcus(ASV0006)and the increased risk of AD.The increased abundance of Streptococcus ASV0006may be a potential modifiable risk factor for AD.
