敲低MALAT1对人鼻咽癌HONE-1细胞增殖、迁移和侵袭的影响OA

目的 探讨敲低人类肺腺癌转移相关转录本 1(metastasis-associated lung adenocarcinoma transcript 1,MALAT1)表达水平对人鼻咽癌HONE-1 细胞增殖、迁移和侵袭的影响.方法 选取人鼻咽癌HONE-1 细胞,将其分为对照组和实验组,两组细胞分别转染control质粒(shcon组)和shMALAT1 质粒(shMALAT1 组),采用荧光显微镜和qRT-PCR法检测两组细胞的转染效率.CCK8 法检测两组细胞的增殖能力,划痕实验检测两组细胞的迁移能力,Transwell法检测两组细胞的迁移和侵袭能力,qRT-PCR法检测两组细胞波形蛋白和上皮钙黏素的mRNA表达水平,Western blot法检测两组细胞波形蛋白和上皮钙黏素蛋白的表达水平.结果 与shcon组相比,敲低MALAT1 后其mRNA的表达水平明显降低,细胞的增殖能力(t=5.520,P<0.000 1)、迁移能力(t=9.285,P<0.000 1;t=9.577,P<0.000 1)及侵袭能力(t=8.481,P<0.000 1)明显降低,波形蛋白mRNA和蛋白的表达水平明显降低,上皮钙黏素mRNA和蛋白的表达水平明显升高.结论 敲低MALAT1 可以抑制人鼻咽癌细胞的增殖、迁移、侵袭和上皮-间充质转化.
Objective To investigate the effect of MALAT1 knockdown on the proliferation and migration and invasion in HONE-1 of nasopharyngeal carcinoma cells.Methods The nasopharyngeal carcinoma HONE-1 cells were divided into two groups,which were transfected with control plasmid(shcon group)and shMALAT1 plasmid(shMALAT1 group)respectively.The transfection rates of the two groups were detected by fluorescence microscope.And MALAT1 gene knockdown level was de-tected by real-time fluorescence quantitative PCR(qRT-RCR).The proliferation abilities were detected by CCK8 method;the migration and invasion ability of the two groups were detected by transwell assay;and the expression levels of vimentin and E-cadherin mRNA were detected by qRT-PCR;and the expression levels of vimentin and E-cadherin protein were detected by West-ern blot.Results Compared with the control group,the expressions level of MALAT1 was significantly decreased after transfection shMALAT1 plasmid.Compared with the control group,the cell proliferation ability(t=5.520,P<0.000 1)and migration a-bility(t=9.285,P<0.000 1;t=9.577,P<0.000 1)and invasion ability(t=8.481,P<0.000 1)in shMALAT1 group was significantly decreased,the expressions level of vimentin mRNA and protein in the shMALAT1 group were significantly de-creased.the expression levels of E-cadherin mRNA and protein in the shMALAT1 group were significantly increased.Conclusion Knocking down MALAT1 gene can inhibit proliferation,migration,invasion and EMT of HONE-1 cells in human nasopharyn-geal cancer.
