以DQ1、DQ8、LS1、金球盖菇、CD1、CD2等6份大球盖菇菌株为试材,采用"25 ℃(对照)→28 ℃→31 ℃→34 ℃"渐进式梯度高温驯化方法,研究了在不同温度下菌丝生长速率、菌丝长势、菌丝干质量、高温胁迫后恢复能力及各菌株耐高温特性,以期突破温度对大球盖菇菌丝生长的限制,筛选适宜高温环境(28~34℃)生长的优良菌株,为高温地区大球盖菇栽培及周年生产体系构建提供参考依据.结果表明:随驯化温度升高,6个菌株的菌丝生长均受抑制,但抑制程度存在显著差异(P<0.05).34 ℃重度高温驯化阶段,DQ1与DQ8菌株表现最优,其菌丝生长速率分别达0.43、0.41 cm·d-1,显著高于其他菌株;菌丝长势维持4级(较浓密、无褐变),菌丝干质量分别为0.82、0.79 g·(100mL)-1;34 ℃胁迫后转移至25 ℃培养24 h,2个菌株菌丝恢复率均达100%.LS1、金球盖菇菌株在31 ℃以上出现菌丝严重褐变、生长停滞,CD1、CD2菌株耐高温能力中等.综上,DQ1与DQ8菌株的耐高温性能突出,可作为高温环境下大球盖菇菌丝培养阶段的优选菌株.

Taking six Stropharia rugosoannulata strains(DQ1,DQ8,LS1,Golden Stropharia,CD1,CD2)as test materials,a progressive gradient high-temperature acclimation method(25℃(control)→28 ℃→ 31 ℃→ 34℃)was employed to study mycelial growth rate,mycelial vigor,mycelial dry mass,post-high-temperature stress recovery capacity,and high-temperature tolerance characteristics of each strain,in order to overcome temperature limitations on Stropharia rugosoannulata mycelial growth,screen superior strains suitable for high-temperature environments(28-34℃),and provide reference for cultivating Stropharia rugosoannulata in high-temperature regions and establishing year-round production systems.The results showed that as acclimation temperature increased,mycelial growth of all six strains was inhibited,but the degree of inhibition varied significantly(P<0.05).During the severe high-temperature acclimation stage at 34℃,strains DQ1 and DQ8 performed the best,with mycelial growth rates reaching 0.43 cm·d-1 and 0.41 cm·d-1,respectively,significantly higher than other strains;mycelial vigor remained at level 4(dense and brown-free),with mycelial dry masses of 0.82 g·(100mL)-1 and 0.79 g·(100mL)-1;after 34℃ stress and transfer to 25℃cultivation for 24 hours,both strains achieved 100%mycelial recovery.Strains LS1 and Golden stropharia exhibited severe mycelial browning and growth stagnation above 31℃,while strains CD1 and CD2 showed moderate high-temperature tolerance.In conclusion,strains DQ1 and DQ8 demonstrated outstanding high-temperature resistance and could be selected as optimal strains for Stropharia rugosoannulata mycelial cultivation in high-temperature environments.
