以酸枣抗病健康叶(KB)和感病叶(ZFB)为试材,采用NaNO2-Al(NO3)3-NaOH显色法测定KB和ZFB的类黄酮含量,转录组测序技术和实时荧光定量PCR技术筛选与抗枣疯病相关的代谢通路和差异表达基因并进行定量验证,研究了枣疯病植原体对酸枣的类黄酮生物合成途径影响,以期为酸枣的品种选育提供参考依据.结果表明:KB vs ZFB共收集到4 682个差异表达基因(DEGs),其中有1 540个基因上调,3 142个基因下调.GO富集分析中,共有4 066个差异表达基因被注释,包括类黄酮生物合成过程、类黄酮代谢过程等.KEGG富集通路中,也显著富集在类黄酮生物合成等通路.根据转录组数据库中4 682个DEGs的注释,鉴定出25个与类黄酮生物合成相关的关键基因,包括HCT、CCoAOMT基因,其在KB vs ZFB组中明显上调表达,能够促进阿魏酸辅酶A的积累;226个转录因子家族基因数,主要集中于ERF、MYB、bHLH等与类黄酮生物合成相关转录因子家族.

Taking jujube witches' disease-resistant healthy leaves(KB)and disease-susceptible leaves(ZFB)as experimental materials,the flavonoid content of KB and ZFB was determined by NaNO2-Al(NO3)3-NaOH colorimetric method.Transcriptome sequencing technology and qRT-PCR technology were used to screen for metabolic pathways and differentially expressed genes related to resistance to jujube disease,and quantitative verification was performed.The effects of jujube witches'-broom phytoplasma on the flavonoid biosynthesis pathway of sour jujube were studied,in order to provide reference for the breeding of sour jujube varieties.The results showed that a total of 4 682 differentially expressed genes(DEGs)were collected between KB and ZFB,of which 1 540 genes were upregulated and 3 142 genes were downregulated.In GO enrichment analysis,a total of 4 066 differentially expressed genes were annotated,including flavonoid biosynthesis and flavonoid metabolism processes.In the KEGG enrichment pathway,it was also significantly enriched in pathways such as flavonoid biosynthesis.Based on the annotations of 4 682 DEGs in the transcriptome database,25 key genes related to flavonoid biosynthesis were identified,including HCT and CCoAOMT genes,which were significantly upregulated in the KB vs ZFB group and could promote the accumulation of ferulic acid coenzyme A.There were 226 transcription factor family genes,mainly concentrated in ERF,MYB,bHLH families known to regulate flavonoid metabolism.
