金锁固精丸加味通过调控Podocin/Nephrin和整合素表达减轻糖尿病肾病足细胞损伤的机制研究OA
Mechanism of Modified Jinsuo Gujing Pill in Attenuating Diabetic Nephropathy Podocyte Injury by Regulating Podocin/Nephrin and Integrin Expression
目的:观察金锁固精丸加味对糖尿病肾病大鼠足细胞损伤的影响,并探讨其通过调控足细胞关键蛋白Podocin、Nephrin及整合素α3、β1表达发挥保护作用的机制.方法:将36只清洁级雄性SD大鼠随机分为正常组6只、模型组15只和金锁固精丸加味干预组15只.采用链脲佐菌素腹腔注射诱导糖尿病肾病模型.造模成功1周后,干预组予金锁固精丸加味方颗粒剂灌胃,正常组与模型组予等量生理盐水,每日灌胃给药1次,持续12周.于第4、12周动态监测血糖、24 h尿蛋白定量及尿足细胞计数.治疗结束后处死大鼠,取肾皮质,采用实时荧光定量PCR法检测Podocin、Nephrin、整合素α3及β1的mRNA表达水平.结果:干预12周后,与模型组比较,干预组血糖水平显著降低(P<0.01),24 h尿蛋白排泄量及尿足细胞数量均显著减少(P<0.01).同时,干预组肾皮质中Podocin、Nephrin、整合素α3及β1的mRNA表达水平较模型组显著上升(P<0.01).模型组上述分子的mRNA表达水平较正常组显著下降(P<0.01).结论:金锁固精丸加味能降低糖尿病肾病大鼠血糖,减少蛋白尿及尿足细胞排泄,其机制可能与上调足细胞裂孔膜关键蛋白(Podocin、Neph-rin)及黏附分子(整合素α3、β1)的mRNA表达,从而保护足细胞、减轻损伤有关.
Objective:To observe the effect of Modified Jinsuo Gujing Pills(MJGP)on podocyte injury in diabetic nephropathy(DN)rats and to explore its mechanism by regulating the expression of key podocyte proteins,Podocin,Nephrin,and integrins α3 and β1.Methods:Thirty-six clean-grade male Sprague-Dawley(SD)rats were randomly divided into a normal group(n=6),a model control group(n=15),and a Modified Jinsuo Gujing Pills intervention group(n=15).The DN model was induced by intraperitoneal injection of streptozotocin(STZ).One week after successful modeling,the intervention group received Modified Jinsuo Gujing Pills granule suspension by gavage,while the normal and model groups received an equal volume of normal saline for 12 consecutive weeks.Blood glucose,24-hour urinary protein,and urinary podocyte count were dynamically monitored at the 4th and 12th weeks.After treatment,rats were sacrificed,and renal cortex tissues were collected.The mRNA expression levels of Podocin,Nephrin,and integrins α3 and β1 were detected by real-time quantitative polymerase chain reaction(qPCR).Results:After 12 weeks of intervention,compared with the model group,the intervention group showed significantly lower blood glucose levels(P<0.01),significantly reduced 24-hour urinary protein excretion and urinary podocyte count(both P<0.01).Concurrently,the mRNA expression levels of Podocin,Nephrin,integrin α3,and integrin β1 in the renal cortex of the intervention group were significantly increased compared to the model group(all P<0.01).The mRNA expression levels of these molecules in the model group were significantly lower than those in the normal group(all P<0.01).Conclusion:Modified Jinsuo Gujing Pills can lower blood glucose,reduce proteinuria and urinary podocyte excretion in DN rats.The mechanism may be related to upregulating the mRNA expression of key slit diaphragm proteins(Podocin,Nephrin)and adhesion molecules(integrins α3,β1)in podocytes,thereby protecting podocytes and alleviating injury.
