肝细胞癌中脂肪酸代谢的研究进展OA
Recent advances in fatty acid metabolism in hepatocellular carcinoma
肝细胞癌(HCC)是全球常见的恶性肿瘤,其发病率和死亡率居高不下,其发病机制涉及多种因素,其中脂质代谢改变起着关键作用.HCC中脂肪酸合成代谢途径显著上调,涉及的关键酶如ATP柠檬酸裂解酶(ACLY)、乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FASN)和硬脂酰辅酶A去饱和酶1(SCD1)在HCC组织中表达上调,促进肿瘤生长和进展.此外,脂肪酸氧化(FAO)在HCC细胞中也至关重要,尤其在葡萄糖剥夺条件下为细胞供能.固醇调节元件结合蛋白(SREBP)和过氧化物酶体增殖物激活受体(PPAR)作为调控因子,影响脂肪酸合成和分解代谢.针对这些酶的抑制策略可能有效阻断肿瘤发展,为癌症治疗提供新方向.因此,本文对脂肪酸代谢相关蛋白质和基因在HCC发生发展中的研究进展进行综述,希望这些发现为针对HCC的脂质代谢靶向治疗提供新思路.
Hepatocellular carcinoma(HCC)is a globally prevalent malignant tumor,characterized by persistently high incidence and mortality rates.The pathogenesis of HCC is multifactorial,with dysregulated lipid metabolism emerging as a critical factor.In HCC,the anabolic pathways of fatty acid synthesis are markedly upregulated,with key enzymes such as ATP-citrate lyase,acetyl-CoA carboxylase,fatty acid synthase,and stearoyl-Coenzyme A desaturase 1 being overexpressed in HCC tissues,thereby facilitating tumor growth and progression.Moreover,fatty acid oxidation plays a vital role in HCC cells,especially under conditions of glucose deprivation,as it serves as an alternative energy source.Sterol regulatory element-binding proteins and peroxisome proliferators-activated receptors function as key regulatory factors that modulate the synthesis and catabolism of fatty acids.Strategic inhibition of these enzymes could effectively arrest tumor development,paving new avenues for cancer therapeutic interventions.Hence,this review synthesizes the research advancements in fatty acid metabolism-related proteins and genes in the occurrence and development of HCC,with the aim of providing novel perspectives for lipid metabolism-targeted therapeutic strategies against HCC.
