槲芪方及其拆方治疗原发性肝癌的机制研究OA
Investigation into the mechanisms of HuQi Formula and its deconstructed groups for Hepatocellular Carcinoma treatment
目的:探究槲芪方及其拆方对肝细胞癌的抗肿瘤活性,初步探讨其作用机制.方法:通过构建肝癌皮下瘤模型,采用灌胃给药方式评价槲芪方的抗肝癌作用;并基于药物功效将其拆分为补肾益气组(槲寄生、生黄芪)、活血化瘀组(丹参、醋莪术)、清热解毒组(白花蛇舌草、叶下珠),探究槲芪方及各拆方组的体内抗肝癌活性;同时收集小鼠外周血和肝癌组织,检测外周血中免疫、炎症相关指标,初步阐明其作用机制.结果:槲芪方及其拆方均能够显著抑制小鼠肝癌皮下移植瘤的生长.活血化瘀组、清热解毒组药物可降低IL-6、IL-17A等炎症相关细胞因子的表达发挥抗肿瘤作用;补肾益气组药物通过升高小鼠外周血中CD4+T细胞、CD8+T细胞和CD8+IFN-γ+T细胞百分比,降低Th2 类细胞因子水平提高机体免疫功能;槲芪方组在此基础上可降低小鼠外周血中与肝癌转移、复发相关指标VEGF的表达.结论:槲芪方及其拆方具有良好的体内抗肿瘤活性,槲芪方能通过提高机体免疫力、降低炎症反应等多途径发挥抗肿瘤作用,复方配伍能提高疗效.
Objective:To investigate the antitumor activity of HuQi Formula and its prescription-disassembled groups against hepatocellular carcinoma(HCC)and preliminarily explore its underlying mechanism.Methods:A subcutaneous xenograft model of HCC was established in mice,and intragastric administration was adopted to evaluate the anti-HCC effect of HuQi Formula.Based on the medicinal efficacies,the formula was divided into three prescription-disassembled groups:the Kidney-Tonifying and Qi-Invigorating Group(Mistletoe,Raw Astragalus),the Blood-Activating and Stasis-Removing Group(Danshenmiltiorrhiza,Curcuma zedoaria),and the Heat-Clearing and Detoxifying Group(Hedentaria diffusa,Phyllanthus urinaria).The in vivo antitumor activity of HuQi Formula and each of its disassembled groups was investigated.Meanwhile,peripheral blood and HCC tissues of mice were collected,and immune-and inflammation-related indicators in peripheral blood were detected to preliminarily elucidate the mechanism of action.Results:HuQi Formula and all its disassembled groups significantly inhibited the growth of subcutaneous HCC xenografts in mice.The Blood-Activating and Stasis-Removing Group and the Heat-Clearing and Detoxifying Group exerted antitumor effects by downregulating the expression of inflammation-related cytokines such as IL-6 and IL-17A.The Kidney-Tonifying and Qi-Invigorating Group enhanced the body's immune function by increasing the percentages of CD4+T cells,CD8+T cells and CD8+IFN-γ+T cells in mouse peripheral blood and reducing the levels of Th2-type cytokines.On this basis,HuQi Formula further decreased the expression of VEGF,an indicator associated with HCC metastasis and recurrence,in mouse peripheral blood.Conclusion:HuQi Formula and its prescription-disassembled groups exhibit favorable in vivo antitumor activity against HCC.HuQi Formula exerts its antitumor effects through multiple pathways including enhancing immune function and alleviating inflammatory response,and the compatibility of the compound formula can improve the therapeutic effect.
