基于宏基因组学探讨扶正消癌Ⅰ号方对结直肠癌荷瘤裸鼠肠道菌群的影响OA

Metagenomics-Based Study on Effects of Fuzheng Xiao'ai Formula I on Intestinal Flora in Colorectal Tumor-Bearing Nude Mice

目的:从肠道菌群角度研究扶正消癌Ⅰ号方对结直肠癌(CRC)裸鼠肿瘤的抑制作用.方法:构建 CRC 裸鼠模型,将造模成功的20 只裸鼠随机分为模型对照组和扶正消癌Ⅰ号方低(17.47 g/kg)、中(34.94 g/kg)、高(69.89 g/kg)剂量组,每组5 只.模型对照组灌胃生理盐水,扶正消癌Ⅰ号方各剂量组灌胃对应浓度水煎液,连续干预2 周后处死裸鼠,完整剥离瘤体并取部分肿瘤组织进行 HE 染色.基于瘤重、体积及病理结果筛选出疗效较好的中剂量组,收集该组及模型对照组的盲肠内容物进行宏基因组测序分析.结果:扶正消癌Ⅰ号方各剂量组裸鼠的瘤重和体积均下降(P<0.05,P<0.01).HE 染色显示扶正消癌Ⅰ号方低、中、高剂量组的肿瘤细胞密度降低,且伴有不同程度坏死.宏基因组学测序显示,与模型对照组相比,扶正消癌Ⅰ号方中剂量组的副萨特氏菌属、双歧杆菌属、粪杆菌属、耳蜗形梭菌、啮齿粪杆菌丰度显著增加(P<0.05),变形杆菌属、柠檬酸杆菌属、摩根菌属、肠杆菌属、摩氏摩根菌、霍氏肠杆菌的丰度显著降低(P<0.05).京都基因与基因组百科全书(KEGG)分析发现淀粉与蔗糖代谢通路在两组中差异显著(P<0.05),其中 α-葡萄苷酶、UTP-葡萄糖-1-磷酸尿苷酸转移酶、磷酸葡萄糖变位酶、麦芽糖 α-D-葡萄糖基转移酶在该通路中明显下调(P<0.05).结论:扶正消癌Ⅰ号方通过增加有益菌的丰度,降低有害菌的丰度,进而修复肠道屏障,维持肠道内环境的稳态,而下调淀粉和蔗糖代谢通路中的α-葡糖苷酶可能是其发挥抗CRC的潜在途径.

Objective:To study the inhibitory effect of Fuzheng Xiao'ai Formula I on colorectal cancer tumors in nude mice from the perspective of intestinal flora.Methods:The CRC nude mouse model was constructed and 20 successfully modeled nude mice were randomLy divided into the model control group and low dose(17.47 g/kg),medium dose(34.94 g/kg),and high dose(69.89 g/kg)groups of Fuzheng Xiao'ai Formula I,with 5 mice in each group.The model control group was gavaged with normal saline,and each dose of Fuzheng Xiao'ai Formula I was gavaged with the corresponding concentration of decoction.After 2 consecutive weeks of intervention,the nude mice were sacrificed,the tumor bodies were completely dissected,and some tumor tissues were taken for HE staining.Based on the tumor weight,volume and pathological results,the medium dose group with better efficacy was screened out.Meanwhile,the cecal contents of this group and the model control group were collected for metagenomic sequencing analysis.Results:The tumor weight and volume of nude mice in each dose group of Fuzheng Xiao'ai Formula I decreased.HE staining showed that the density of tumor cells in the low,medium,and high dose groups of Fuzheng Xiao'ai Formula I decreased,accompanied by different degrees of necrosis.Metagenomic sequencing showed that compared with the model control group,the abundances of Parabacteroides,Bifidobacterium,Faecalibacterium,Clostridium auriscati,and Faecalibaculum rodentium in the medium dose group significantly increased(P<0.05),and the abundances of Proteus,Citrobacter,Morganella,Enterobacter,Morganella morganii,and Enterobacter hormaechei significantly decreased(P<0.05).Kyoto Encyclopedia of Genes and Genomes analysis found that the starch and sucrose metabolism pathway was significantly different between the two groups(P<0.05),among which α-glucosidase,UTP-glucose-1-phosphate uridylyltransferase,phosphoglucomutase,and maltose α-D-glucosyltransferase were significantly downregulated in this pathway(P<0.05).Conclusion:Fuzheng Xiao'ai Formula I repairs the intestinal barrier and maintains the homeostasis of the intestinal environment by increasing the abundance of beneficial bacteria and reducing the abundance of harmful bacteria.Down-regulating α-glucosidase in the starch and sucrose metabolism pathway may be a potential way for it to exert anti-colorectal cancer effects.
