阿霉素诱导小鼠急性心肌损伤模型的动态研究及心血康的保护作用OA
Temporal dynamics of Doxorubicin-induced acute myocardial injury in mice and the protective role of Xinxuekang
目的:阐明阿霉素(Doxorubicin,DOX)诱导小鼠急性心肌损伤的时序性病理演变特征与核心毒理机制,明确心血康(XXK)对DOX所致急性心肌损伤的药理学效应,为开发DOX急性心脏毒性的防治药物与干预策略提供实验依据和理论支撑.方法:给小鼠腹腔注射DOX建立急性心肌损伤模型,检测心肌酶、HE染色、透射电镜观察及转录组测序,研究DOX急性心肌损伤特征及心血康的保护作用.结果:DOX组小鼠血清CK-MB和LDH活性在第2天显著升高;小鼠心脏第2天出现轻微空泡和炎症细胞浸润,第4和第6天局部心肌纤维紊乱、细胞间质水肿,第8天有明显间质水肿、胞质空泡化和细胞核固缩及碎裂;第4和第8天,心肌出现线粒体肿胀、嵴结构破坏和细胞膜破坏等损伤.转录组分析表明,心肌损伤早期主要为细胞应激反应,后期则进入修复和适应阶段.心血康干预可有效降低心肌酶活性、保护线粒体结构、减轻心脏病理损伤.结论:DOX诱导的心肌损伤早期为细胞水肿、线粒体功能崩溃和氧化应激反应,后期进入修复和适应阶段.采用心血康干预DOX抗肿瘤治疗前期,对DOX所致急性心肌损伤具有一定保护作用.
Objective:This study aims to elucidate the temporal pathological progression and key toxicological mechanisms of Doxorubicin(DOX)-induced acute myocardial injury in mice,and to evaluate the pharmacological effects of Xinxuekang(XXK),thereby providing an experimental basis for the prevention and treatment of DOX-induced acute cardiotoxicity.Methods:An acute myocardial injury model in Kunming mice through intraperitoneal injection of DOX and assessed cardiac injury was es-tablished.Enzyme assay,HE staining,TEM,and transcriptome profiling were performed to dynamically assess myocardial injury and the effects of XXK.Results:DOX markedly increased serum CK-MB and LDH levels on day 2.Histopathology revealed early vacuolisation and inflammatory infiltration,progressing to fibre disarray and interstitial oedema on days 4 and 6,and marked cytoplasmic vacuolisation with nuclear pyknosis and fragmentation on day 8.TEM demonstrated progressive mitochondrial swell-ing,cristae disruption,and membrane injury on day 4 and 8.Transcriptomic analysis indicated activation of cellular stress path-ways in the early phase,followed by repair and adaptive responses.XXK administration mitigated enzyme elevation,preserved mitochondrial integrity,and alleviated pathological damage.Conclusion:Early DOX-induced cardiotoxicity is characterised by cellular oedema,mitochondrial dysfunction,and oxidative stress,whereas later stages reflect compensatory repair.Early XXK in-tervention confers significant protection against acute DOX-induced myocardial injury.
