灸药联合对三阴性乳腺癌小鼠肿瘤组织抗原提呈细胞CD80及CD86表达的影响OA
Effects of moxibustion combined with medication on CD80 and CD86 expression in tumor tissue antigen-presenting cells of mice with triple-negative breast cancer
目的:观察灸药联合对三阴性乳腺癌(TNBC)小鼠肿瘤组织中抗原提呈细胞(APCs)表面共刺激分子CD80及CD86的表达及血清中CD4+和CD8+T细胞的影响,探讨直接灸联合化疗药物对TNBC的干预效果.方法:将50只BALB/c小鼠根据随机数字表法分为正常组、模型组、药物组、直接灸组和灸药联合组,每组10只.正常组不给予任何干预,其余4组采用4T1细胞注射于小鼠左侧第四乳腺脂肪垫下的方法制备荷瘤小鼠模型.造模结束后1周,直接灸组接受直接灸足三里;药物组接受腹腔注射阿霉素;灸药组同时接受直接灸足三里和腹腔注射阿霉素,共干预21 d.干预结束后测量各组小鼠肿瘤体积;苏木素-伊红染色观察肿瘤组织形态;免疫组化和免疫印迹法检测肿瘤组织中CD80及CD86表达;流式细胞术检测肿瘤组织中CD80阳性表达及外周血中CD4+、CD8+及CD4+/CD8+.结果:与正常组比较,其余组别外周血中CD4+/CD8+均下降(P<0.01).与模型组比较,其余各组肿瘤体积缩小(P<0.01),肿瘤细胞形态异常或碎裂,细胞排列紊乱,CD80、CD86表达不同程度升高(P<0.01或P<0.05),CD4+/CD8+升高(P<0.01).与药物组比较,单独直接灸干预组CD80、CD86表达程度下降(P<0.01或P<0.05),CD4+/CD8+降低(P<0.01),灸药组小鼠上述指标改善效果更显著(P<0.01或P<0.05).结论:与化疗药、直接灸单独干预比较,灸药联合对TNBC小鼠的干预效应更好,在抑制肿瘤生长,上调APCs共刺激分子CD80、CD86的表达,促进T淋巴细胞亚群恢复平衡方面效果更优.
Objective:To observe the effect of moxibustion combined with chemotherapy on the expression of costimulatory molecules CD80 and CD86 on antigen-presenting cells(APCs)in tumor tissues of mice with triple-negative breast cancer(TNBC),and to explore the intervention effect of direct moxibustion combined with chemotherapeutic drugs on TNBC. Methods:Fifty BALB/c mice were divided into a normal group,a model group,a medication group,a direct moxibustion group,and a moxibustion plus medication group using the random number table method,with 10 mice in each group.The normal group was not given any intervention,and the other four groups were injected with 4T1 cells under the left fourth mammary fat pad to prepare tumor-bearing mouse models.One week after modeling,the direct moxibustion group received direct moxibustion at Zusanli(ST36);the medication group received intraperitoneal injection of doxorubicin;the moxibustion plus medication group received direct moxibustion at Zusanli(ST36)and intraperitoneal injection of doxorubicin together,all for 21 d.After the intervention finished,the mouse's tumor volume in each group was measured;hematoxylin-eosin staining was used to observe the morphology of tumor tissues;immunohistochemistry and Western blotting were used to detect the expression of CD80 and CD86 in tumor tissues.The positive expression of CD80 in tumor tissues and CD4+,CD8+,and CD4+/CD8+in peripheral blood was detected by flow cytometry. Results:CD4+/CD8+in the peripheral blood in the other groups were decreased compared to the normal group(P<0.01).Tumor volumes were decreased(P<0.01),cell morphology was abnormal or fragmented,cell arrangement was disordered,the expression of CD80 and CD86 was increased to different degrees(P<0.01 or P<0.05),and CD4+/CD8+was increased(P<0.01)in tumor cells in the other groups compared to the model group.Compared to the medication group,the expression levels of CD80 and CD86(P<0.01 or P<0.05),as well as CD4+/CD8+(P<0.01)were decreased in the direct moxibustion group,and the improvement effect on the above indicators in the moxibustion plus medication group was more significant(P<0.01 or P<0.05). Conclusion:Compared to the chemotherapeutic drug or direct moxibustion alone,the combination of moxibustion with the chemotherapeutic drug has a better intervention effect on TNBC mice,especially in inhibiting tumor growth,upregulating the expression of costimulatory molecules CD80 and CD86 on APCs,and promoting the restoration of T lymphocyte subsets to balance.
