泰山磐石散加减方体外抗氧化活性评价OA

为了评价并探究泰山磐石散加减方的体外抗氧化活性,本试验通过4种经典的体外抗氧化模型(DPPH·、ABTS+·、·OH、FRAP),以维生素C(VC)为阳性对照,测定泰山磐石散加减方对DPPH·、ABTS+·、·OH等自由基的清除活性及Fe3+还原能力(FRAP法),分别在517 nm、734 nm、520 nm和593 nm波长处测定吸光度值,并计算该方剂对3种自由基的清除率,通过FeSO4标准曲线计算该方剂Fe3+还原能力.结果表明,泰山磐石散加减方对3种自由基的清除率均随着药物浓度的升高而增强,表现出明显的效量关系,其IC50值分别为DPPH·0.274 5 mg/mL、ABTS+·0.153 1 mg/mL、·OH 0.526 7 mg/mL,清除活性均低于维生素C(IC50分别为0.002 1 mg/mL、0.011 3 mg/mL、0.024 9 mg/mL);泰山磐石散加减方对Fe3+的还原能力与药物浓度呈现正相关,但该方剂对Fe3+还原能力(0.059 5 mmol/g)显著低于维生素C(100.830 8 mmol/g).上述结果提示,泰山磐石散加减方对DPPH•、ABTS+•、•OH等自由基具有清除作用,并对铁离子具有还原能力.

To evaluate the antioxidant activity of the modified Taishan Panshi San formula in vitro,four classical models(DPPH·,ABTS+·,·OH and FRAP)were employed,with vitamin C(VC)acting as the positive control,to determine its capacity to scavenge DPPH·,ABTS+·,·OH radicals and reduce Fe3+(FRAP assay).Absorbance was measured at 517 nm,734 nm,520 nm and 593 nm,respectively,and the scavenging rates were calculated.Fe3+reduction capacity was determined via a FeSO4 standard curve.The results showed that the scavenging rates of the modified Taishan Panshi San formula against all three free radicals increased with concentration,exhibiting a clear dose-response relationship.The IC50 values were 0.274 5 mg/mL for DPPH·radicals,0.153 1 mg/mL for ABTS+·radicals and 0.526 7 mg/mL for·OH,all of which were lower than the IC50 value of vitamin C(0.002 1 mg/mL,0.011 3 mg/mL and 0.024 9 mg/mL,respectively).The Fe3+reduction capacity of the modified Taishan Panshi San formula exhibited a positive correlation,increasing with formula concentration,while Fe3+reduction capacity of the formulation(0.059 5 mmol FeSO4/g)was significantly lower than that of vitamin C(100.830 8 mmol FeSO4/g).Taishan Panshi San demonstrates scavenging activity against DPPH·,ABTS+·and hydroxyl radicals,as well as iron ion reduction capacity.
