正交试验优选石榴皮最佳炮制工艺研究OA
Optimization of the processing technology for pomegranate peel by orthogonal experiment
本研究旨在通过优化石榴皮加辅料炒制工艺,提高其药效成分含量,为石榴皮炮制规范化提供科学依据.通过预试验对清炒、土炒及麸炒3种常用炒制方法进行比较,以总没食子酸和鞣花酸含量为主要评价指标,初步确定麸炒法为石榴皮最优炒制方法.在此基础上,为进一步优化麸炒工艺,采用正交试验设计,选取炮制时间和麦麸用量作为关键考察因素,每个因素设定3个水平.以炮制后石榴皮中总没食子酸及鞣花酸的含量作为评价指标,通过综合评分法对工艺参数进行分析与优化.结果表明,不同炮制时间与麦麸用量对石榴皮中目标成分含量具有显著影响.通过极差分析与方差分析,确定各因素对综合评分的影响主次顺序为炮制时间>麦麸用量.最终优化得到的最佳工艺条件为炮制时间3 min,麦麸用量20%.所获最佳工艺操作简便、稳定可行,不仅有助于提高石榴皮的药效物质基础,也为推动其炮制工艺标准化、生产规范化提供了可靠的数据支持与理论依据,对促进石榴皮在兽医临床中的合理应用具有积极意义.
This study aimed to optimize the frying process of pomegranate peel with auxiliary materials to increase the content of its bioactive components,providing a scientific basis for standardizing the processing of pomegranate peel.Preliminary experiments were conducted to compare three common frying methods:plain frying,soil frying,and bran frying,using the contents of total gallic acid and ellagic acid as the main evaluation indicators.The results preliminarily identified bran frying as the optimal method for pomegranate peel.Subsequently,to further optimize the bran frying process,an orthogonal experimental design was adopted,with processing time(A)and wheat bran dosage(B)as the key variables,each set at three levels.The contents of total gallic acid and ellagic acid in the processed pomegranate peel were used as evaluation indices,and the process parameters were analyzed and optimized using a comprehensive scoring method.The results showed that both processing time and wheat bran dosage significantly influenced the content of target components in pomegranate peel.Based on range analysis and analysis of variance,the order of influence of each factor on the comprehensive score was determined as follows:processing time had a greater impact than wheat bran dosage.The optimal processing conditions were determined to be a processing time of three minutes and a wheat bran dosage of twenty percent.The optimized process is simple,stable,and feasible.It not only helps enhance the material basis for the efficacy of pomegranate peel,but also provides reliable data support and a theoretical foundation for promoting the standardization and normative production of its processing technology.This study holds positive implications for advancing the rational application of pomegranate peel in clinical practice and pharmaceutical preparations.
