根际土壤微生物与植物生长发育及产量品质紧密相关.为更充分发掘药用植物根际功能芽孢杆菌资源,本研究以甘草根际土壤为研究对象,分离并纯化了5株芽孢杆菌;形态鉴别、生理生化特征分析及16S rRNA分子鉴定表明,G1-a菌株为海恩西芽孢杆菌,G1-E、G5-1、G5-2菌株为萎缩芽孢杆菌,G6-f菌株为环状芽孢杆菌,同时检测各菌株产酶和生防功能.G5-1、G6-f、G1-E菌株具有产淀粉酶和蛋白酶功能,G5-1、G6-f及G5-2菌株还具备较强解钾、溶磷功能以及抑制病原菌活性,G6-f菌株对木贼镰刀菌、接骨木镰刀菌和茄病镰刀菌的抑菌活性最强,5株芽孢杆菌均具备固氮、产生物膜和分泌IAA潜能,G5-1、G5-2、G1-a菌株还可分泌铁载体;分离菌株均可在2%~10%NaCl、pH 4~10、温度4℃~48℃范围内生长,G5-1、G5-2、G6-f菌株对盐碱及高温环境具有较强的耐受性.上述结果提示,5株芽孢杆菌在产酶、溶磷、拮抗、固氮、耐盐碱、抑制病原菌等方面具有良好性能,为中药材微生物肥料和生防制剂的制备及应用提供了优质的菌种资源和理论依据.

Rhizosphere soil microorganisms are closely linked to plant growth,development,yield,and quality.To better exploit the functional Bacillus resources in the rhizosphere of medicinal plants,five Bacillus strains from the rhizosphere soil of Glycyrrhiza uralensis Fisch.were isolated and purified.Identification based on morphological,physiological,biochemical characteristics,and 16S rRNA gene sequencing revealed that strain G1-a was Bacillus haynesii,strains G1-E,G5-1,and G5-2 were Bacillus atrophaeus,and strain G6-f was Bacillus circulans,with each strain further assessed for enzyme production and biocontrol potential.Strains G5-1,G6-f,and G1-E demonstrated the ability to produce amylase and protease,while strains G5-1,G6-f,and G5-2 also exhibited strong capabilities in solubilizing potassium,dissolving phosphorus,and inhibiting pathogens.Strain G6-f showed the most effective antagonistic activity against Fusarium equiseti,Fusarium sambucinum,and Fusarium solani.All five Bacillus strains displayed potential for nitrogen fixation,biofilm formation,and indole-3-acetic acid(IAA)secretion,with G5-1,G5-2,and G1-a additionally capable of producing siderophores.The isolated strains were able to grow under conditions of 2%-10%NaCl,pH 4-10,and temperatures ranging from 4℃to 48℃,among which G5-1,G5-2,and G6-f exhibited strong tolerance to saline-alkaline and high-temperature environments.In summary,these five Bacillus strains demonstrate excellent properties in enzyme production,phosphorus solubilization,antagonism,nitrogen fixation,saline-alkaline tolerance,and pathogen inhibition.This study provides high-quality strain resources and a theoretical foundation for the development of microbial fertilizers and biocontrol agents for Chinese herbal medicine.
