31种中药提取液对猪链球菌体外抑菌活性及复方筛选研究OA
The antibacterial activity of 31 Chinese herbal medicine extracts against Streptococcus suis in vitro and screening of their compound formulations
本试验旨在筛选对猪链球菌(Streptococcus suis)具有良好抑菌效果的复方中药.分别采用蒸馏法及浸提法对31味中药进行提取,采用牛津杯法、二倍稀释法测定各中药提取物对猪链球菌的体外抑菌圈大小、最低抑菌浓度(MIC)及最小杀菌浓度(MBC),选取其中抑菌活性较强的4味中药,按照不同的组合进行1∶1的复配,并测定复配后各组中药的MIC及MBC值,筛选最佳的抗猪链球菌中药复方组合.结果显示,在供试的31种中药浸提液及蒸馏液中,对猪链球菌具有抑菌作用的有18种,苏木、黄连、诃子、五倍子、黄柏、大蒜、黄芩等中药浸提液及桂皮蒸馏液的抑菌圈直径均大于20 mm,为高度敏感;五倍子、黄连、苏木、黄柏等中药浸提液的MIC与MBC分别为0.49 mg/mL与0.98 mg/mL、0.98 mg/mL与0.98 mg/mL、0.98 mg/mL与1.95 mg/mL、1.95 mg/mL与3.90 mg/mL,均小于其他处理组;将上述4种中药浸提液列为11个组合进行复配,苏木与五倍子配伍组的MIC、MBC均为0.24 mg/mL,小于其他处理组.体外抑菌试验结果表明,在31味药材中,苏木、黄连、黄柏、五倍子对猪链球菌的体外抑菌活性较强;在复方组合中,苏木与五倍子配伍体外抑菌活性最强,其MIC与MBC值均最低.
The purpose of this experiment is to screen compound Chinese medicines with good antibacterial effects on Streptococcus suis. 31 traditional Chinese medicines were extracted using distillation and extraction methods,respectively. The Oxford cup method and double dilution method were used to determine the in vitro antibacterial zone size,minimum inhibitory concentration (MIC),and minimum bactericidal concentration (MBC) values of each traditional Chinese medicine extract against Streptococcus suis. The four traditional Chinese medicines with the strongest antibacterial effect were selected and compounded in different combinations in a 1:1 ratio. The MIC and MBC values of each group of traditional Chinese medicines after compounding were measured to screen for the best anti Streptococcus suis traditional Chinese medicine compound combination. The results showed that among the 31 traditional Chinese medicine extracts and distillates tested,18 had antibacterial effects on Streptococcus suis. In the inhibition zone test,the diameters of the inhibition zones of Sappan Lignum,Coptidis Rhizoma,Chebulae Fructus,Galla Chinensis,Phellodendri Chinensis Cortex,Allii Sativi Bulbus,Scutellariae Radix extract,and Cinnamomi Cortex distillate were all greater than 20 mm,indicating high sensitivity. In the MIC and MBC determination experiments,the MIC and MBC of the extracts of Galla Chinensis,Coptidis Rhizoma,Sappan Lignum,Phellodendri Chinensis Cortex were 0.49 mg/mL and 0.98 mg/mL,0.98 mg/mL and0.98 mg/mL,0.98 mg/mL and 1.95 mg/mL,and 1.95 mg/mL and 3.90 mg/mL,respectively,which were lower than those of other treatment groups. The extracts of these 4 Chinese herbal medicines were grouped into 11 combinations and compounded. After compounding,the MIC and MBC of the combination of Sappan Lignum and Galla Chinensis was both0.24 mg/mL,which was lower than those of other treatment groups. The results of the in vitro antibacterial test showed that among the 31 medicinal herbs,the single herb of Sappan Lignum,Coptidis Rhizoma,Phellodendri Chinensis Cortex,and Galla Chinensis present the best in vitro antibacterial effect on Streptococcus suis. In the compound combination,the combination of Sappan Lignum and Galla Chinensis has the strongest in vitro antibacterial activity with the lowest MIC and MBC values.
