目的 基于网络药理学和实验验证探讨黄芪-丹参-三七角药对心力衰竭模型大鼠氧化应激的影响.方法 通过网络药理学筛选黄芪-丹参-三七角药防治心力衰竭氧化应激的药物活性成分、核心靶点和信号通路,并将核心靶点与药物活性成分进行分子对接.将40只SD大鼠随机分为4个组:对照组、模型组、角药组和西药组,按照分组给予相应的药物干预.利用心脏彩超比较各组心功能指标,利用HE染色比较各组心脏组织病理改变,Masson染色比较各组心肌细胞纤维化情况;透射电镜观察各组心肌细胞线粒体超微结构变化;比较各组大鼠心肌组织中氧化应激指标.结果 筛选到关键药性成分有槲皮素、山奈酚、木犀草素,核心作用靶点有TNF、IL6、IL1B、TP53、AKT、CASP3.动物实验结果显示,与模型组比较,大鼠心功能明显改善,HE和Masson染色结果显示,心肌组织结构损伤减轻,心肌细胞纤维化减少,电镜结果中可见线粒体结构损伤减轻,同时心肌组织中ROS水平降低,CAT、T-AOC水平升高(P<0.05).结论 角药能够通过减轻氧化应激,减轻线粒体结构和功能损伤,促进心脏收缩舒张功能的恢复,从而对心力衰竭发挥保护作用.

Aim To investigate the effects of Astragalus-Danshen-Sanqi horn medicine on oxidative stress in rats with heart failure model based on network pharmacology and experimental validation.Methods The drug active components,core targets and signal-ing pathways of Astragalus-Danshen-Sanqi horn medi-cine against oxidative stress in heart failure were screened by network pharmacology,and the core tar-gets were molecularly docked with the drug active com-ponents.Forty SD rats were randomly divided into four groups,the control group,model group,horn medi-cine group and western medicine group,and the corre-sponding drug interventions were given according to the subgroups.The changes in the level of cardiac function indexes in each group were compared using cardiac ultrasound,the pathological changes in car-diac tissues in each group were evaluated using HE staining,and the fibrosis of cardiomyocytes in each group was assessed using Masson staining;the changes in the ultrastructure of mitochondria of cardio-myocytes in each group were observed using transmis-sion electron microscopy;and the changes in the level of oxidative stress indexes in the cardiac tissues of rats in each group were compared.Results The key phar-macological components screened were quercetin,kaempferol,and lignocerotoxin,and the core targets of action were TNF,IL6,IL1B,TP53,AKT,and CASP3.Animal experiments showed that the cardiac function of rats was significantly improved when com-pared with that of the model group,and the results of HE and Masson staining showed that the structural damage of myocardial tissues was attenuated,the car-diomyocytes were less fibrous.Electron microscopy re-vealed reduced mitochondrial structure damage,while ROS levels in myocardial tissue were reduced,and CAT and T-AOC levels increased(P<0.05).Conclu-sion Horn medicine is able to exert a protective ef-fect against heart failure by reducing oxidative stress,mitigating mitochondrial structural and functional dam-age,and promoting the restoration of cardiac systolic-diastolic functions.
