二倍体和四倍体南川百合差异性状的转录组分析OA
Transcriptome Analysis of Diffeential Taits Between Diploid and Tetrapllid Lilium rosthorii
为探究南川百合(Lilium rosthornii)四倍体与二倍体性状差异的原因,对2种倍性南川百合叶片的淀粉和可溶性糖含量进行测定,并进一步对其进行转录组测序分析.结果发现,南川百合同源四倍体叶片的可溶性糖及淀粉含量均显著高于二倍体,且随生长发育进程差异显著.与二倍体相比,四倍体转录组测序共发现20 441个差异表达基因,其中有5 006个差异表达基因被注释到GO(gene ontology)条目,主要富集在植物生长、信号转导调控等功能中;KEGG(Kyoto encyclopedia of genes and genomes)富集分析发现,差异表达基因在外来物质的代谢、玉米素合成、氨基酰基tRNA合成等途径差异显著.此外,筛选出7个与糖代谢相关的差异表达基因,富集在UDP葡萄糖基转移酶活性、O-糖基化合物水解酶活性功能;还筛选出10个与病原菌响应相关的差异表达基因,富集在植物-病原体相互作用、植物MAPK信号通路.以上研究结果为南川百合基因功能的深入研究提供数据基础,为推动百合多倍化进程及优良种质资源培育提供理论依据和数据支持.
To investigate the changes of Lilium rosthorii homologous tetraploid with diploid,the contents of starch and soluble sugar in leaves of two kinds of ploidies of L.rosthorii were determined,and transcriptome sequencing analysis were performed.The results showed that the contents of soluble sugar and starch in leaves of the homologous tetraploid of L.rosthorii were significantly higher than those of the diploid,and the differences were significant with the extension of growth and development process.Compared with diploid,a total of 20 441 differentially expressed genes(DEGs)were found in tetraploid plants,among which 5 006 DEGs were annotated in GO(gene ontology)database,mainly enriched in plant growth,signal transduction regulation and other functions,and KEGG(Kyoto encyclopedia of gene and genomes)pathway analysis revealed significant differences in differential genes in pathways such as foreign substance metabolism,zeaxanthin synthesis and aminoacyl tRNA synthesis.In addition,7 DEGs related to glucose metabolism were screened and enriched in UDP glucosyltransferase activity and O-glycosyl compound hydrolase activity,and 10 DEGs related to pathogen response were screened and enriched in plant pathogen interactions and plant MAPK signaling pathway.Above results provided data foundation for in-depth research on gene function,also provided theoretical basis and data support for promoting the polyploidization process of lilies and cultivating excellent germplasm resources.
