Analysis of Genetic Diversity Between Low-molecular-weight Glutenin Subunits and Grain Quality in Newly Bred Wheat Varieties(Lines)at the Huanghuai Wheat Region

为明晰黄淮麦区新育小麦品种(系)低分子量麦谷蛋白亚基(low molecular weight-glutenin subunits,LMW-GS)与籽粒品质的遗传多样性,以近年新审定或正在参试的84份小麦新品种(系)为研究对象,采用22对特异性引物对LMW-GS基因进行检测,同时结合蛋白质含量等6项籽粒品质指标进行综合分析,探索LMW-GS与品质的关系.结果表明,共检测到9种LMW-GS等位基因,其中Glu-A3位点检出A3d(58.33%)、A3a/c(47.62%)和 A3a(4.76%)3 种类型;Glu-B3 位点仅检出 B3d,频率为 29.76%;Glu-D3 位点检出 D3-21(69.05%)、D3-32(58.33%)、D3-23(41.67%)、D3-31(2.38%)和D3-22(1.19%)5种类型.A3a/c和B3d与湿面筋含量呈显著正相关.LMW-GS 组合类型共 40 种,频率较高的为 A3d+D3-21(17.86%)、A3d+D3-23+D3-32(7.14%)和A3a/c+D3-21(7.14%).在优质小麦品种中,Glu-B3d和Glu-D3-32的频率较高.依据籽粒品质指标将84份品种(系)划分为4个类群,其中类群Ⅱ在6项品质指标上表现较好;类群Ⅳ聚集了新麦26、西农979、郑麦366等强筋或中强筋的品种(系).研究结果为小麦品质育种工作提供了重要的理论参考.

To clarify the grain quality and genetic diversity of low-molecular-weight glutenin subunits(LMW-GS)in newly bred wheat varieties(lines)at the Huanghuai wheat region,84 newly approved or under trial wheat varieties(lines)were used as materials.The composition and frequency distribution of LMW-GS were detected using 22 pairs of specific primers,and cluster analysis was conducted in combination with 6 grain quality indicators.The results showed that a total of 9 LMW-GS alleles were detected.At the Glu-A3 locus,3 alleles were detected including Glu-A3d(58.33%),Glu-A3a/c(47.62%)and Glu-A3a(4.76%).At the Glu-B3 locus,only the Glu-B3d allele was detected,with a frequency of 29.76%.At the Glu-D3 locus,Glu-D3-21(69.05%),D3-32(58.33%),D3-23(41.67%),D3-31(2.38%)and D3-22(1.19%)alleles were detected.Both A3a/c and B3d showed a significant positive correlation with wet gluten content.A total of 40 LMW-GS combination types were identified,and the freqnencies of A3d+D3-21(17.86%),A3d+D3-23+D3-32(7.14%)combinations were high.Based on grain quality traits,84 varieties(lines)were clustered into 4 groups.Group Ⅱ exhibited superior comprehensive quality characteristics.Group Ⅳ included multiple strong-gluten or medium-strong-gluten varieties,such as Xinmai 26,Xinong 979 and Zhengmai 366,etc.Above results provided important theoretical reference for wheat quality breeding.
