Genetic diversity analysis of 38 raspberry resources in Qiandongnan of Guizhou

为探究黔东南树莓资源的亲缘关系,了解其遗传距离远近,进而为后续树莓良种认定、品种选育及合理选配亲本提供参考,以黔东南州38份树莓资源为材料,基于SRAP分子标记技术分析其遗传多样性.结果表明,从80对引物中筛选出10对具有多态性的引物组合,这10对引物扩展出的条带多态性比率为96.20%;黔东南野生树莓资源和栽培品种两个居群的观测等位基因数为1.962 4,有效等位基因数为1.335 1,Nei's基因多样性指数为0.219 6,Shannon信息指数为0.356 2,野生资源的有效基因数和基因多样性指数(H)、Shannon's信息指数(Ⅰ)均小于栽培品种;聚类分析中,栽培品种与野生资源没有明显分开聚类,插田藨资源与其他37份资源遗传距离较大.SRAP分子标记在对树莓的遗传多样性研究中具有重要价值,未来可对插田藨类树莓进行更加深度的基因研究,为树莓育种工作提供更多更准确的信息.

To explore the genetic relationships and determine the genetic distances among raspberry resources in Qiandongnan,so as to provide a scientific basis for elite cultivar identification,variety breeding,and rational parent selection,a total of 38 raspberry accessions from Qiandongnan Prefecture were used as materials to analyze their genetic diversity using sequence-related amplified polymorphism(SRAP)molecular marker technology.The results showed that 10 polymorphic primer combinations were selected from 80 primer pairs,and the polymorphism rate of the bands amplified by these 10 primer combinations was 96.20%.The 38 wild and cultivated raspberry accessions were divided into two populations,with an observed allele number of 1.9624 and an effective allele number of 1.335 1.At the same time,the Nei's gene diversity index was 0.219 6 and the Shannon information index was 0.356 2.The effective gene number,gene diversity index(H),and Shan-non's information index(Ⅰ)values of wild raspberry resources were all lower than those of cultivated varieties.The results of cluster analysis showed that cultivated varieties and wild germplasm are not clustered separately,and the genetic distance between Rubus coreanus(Miq.)Miq.,and the other 37 accessions was relatively large.SRAP molecular markers are highly valuable for studying the genetic diversity of raspberries.Future research should focus on in-depth genetic studies of R.coreanus to provide more accurate information for raspberry breeding programs.
