决明子提取物和多烯磷脂酰胆碱对小鼠酒精性肝损伤的影响OA
Effects of Cassia seed extract and polyene phosphatidylcholine on alcohol liver injury in mice
目的 探讨决明子提取物(CSE)与多烯磷脂酰胆碱(PPC)对小鼠酒精性肝损伤的干预作用.方法 40 只C57BL/6J 小鼠按体质量均衡分为空白对照组、模型对照组、CSE 组、PPC 组、CSE 和 PPC 联合组,采用 NIAAA 模型(酒精液体饲料喂养 10 天+单次大剂量酒精灌胃)建立 C57BL/6J 小鼠的肝损伤模型,分别给予 CSE(2 g·kg-1·d-1)、PPC(200 mg·kg-1·d-1)及联合干预.检测体重、摄食量、肝指数,分析血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、甘油三酯(TG)及肝组织超氧化物歧化酶(SOD)水平,观察肝脏病理变化.结果 与空白对照组相比,模型对照组小鼠摄食量(P<0.001)、体重明显降低(P<0.05),肝指数明显升高(P<0.01),血清 ALT(P<0.001)、AST(P<0.05)、TG(P<0.01)显著升高,肝组织 SOD 有所降低,肝脂肪变性明显.干预后 PPC 组 ALT(P<0.001)和 TG(P<0.05)水平显著降低;CSE 组 ALT 水平降低(P<0.05)且 SOD 活性升高(P<0.01);联合组(n=6)较CSE 组 ALT、AST 降低(P<0.05),SOD 有升高趋势,摄食量改善,且病理显示脂肪变性改善最显著.结论 CSE 与PPC 通过协同调控氧化应激和脂质代谢,可有效改善酒精性肝损伤,联合用药效果优于单一干预,为酒精性肝病防治提供新策略.
Objective To investigate the intervention effects of Cassia seed extract(CSE)and polyene phosphatidylcholine(PPC)on alcohol liver injury in mice.Methods 40 C57BL/6J mice were randomly divided into blank control group,model control group,CSE group,PPC group,and CSE-PPC combination group.A liver injury model in C57BL/6J mice was estab-lished using the NIAAA model(10 days of alcohol liquid feed+single high-dose alcohol gavage),and mice were subsequently treated with CSE(2 g·kg-1·d-1),PPC(200 mg·kg-1·d-1),and a combination of both,respectively.Serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglycerides(TG),and hepatic superoxide dismutase(SOD)levels were analyzed,and liver pathological changes were also evaluated.Results Compared with the blank control group,the model control group mice showed significantly reduced food intake(P<0.001)and body weight(P<0.05),as well as marked-ly elevated liver index(P<0.01).Serum ALT(P<0.001),AST(P<0.05),and TG(P<0.01)levels significantly in-creased,while liver tissue SOD levels decreased,and hepatic steatosis was pronounced.After intervention,the levels of ALT(P<0.001)and TG(P<0.05)significantly reduced in the PPC group;the ALT level decreased(P<0.05)and SOD activity increased(P<0.01)in the CSE group;the combined group(n=6)showed a reduction in ALT and AST(P<0.05)compared to the CSE group,SOD activity showed a trend of increase,food intake improved,and pathology showed the most significant improvement.Conclusion CSE and PPC can effectively ameliorate alcohol liver injury through synergistic regulation of oxida-tive stress and lipid metabolism.The combined use of these drugs yields better results than single intervention,which provides a new strategy for the prevention and treatment of alcoholic liver disease.
