目的 探究核受体结合 SET 结构域蛋白家族成员(NSD1、NSD2、NSD3)在乳腺癌中的表达差异、功能作用及临床预后价值.方法 利用 Sangerbox、GEPIA 2.0、HPA、Kaplan-Meier Plotter 数据库分析 NSD 家族在乳腺癌中的表达水平及预后价值,使用 cBioPortal 数据库分析 NSD 家族的基因变异情况及生存差异,通过 TIMER3 数据库分析NSD 家族与免疫评分的相关性,使用STRING 数据库及TCGA 数据集分析NSD 蛋白互作网络及与肿瘤信号通路的关联性.结果 乳腺癌中 NSD1、NSD2、NSD3 的 mRNA 和蛋白表达均显著上调,且家族成员间存在表达相关性.NSD1高表达与无远处转移生存期缩短相关,NSD2 高表达与乳腺癌患者无进展生存期(RFS)、总生存期(OS)、无远处转移生存期(DMFS)缩短显著相关,NSD3 高表达患者无进展生存期略长,但总生存期和无远处转移生存期更短,预后更差.NSD 家族各成员基因变异率为 1.3%~12.4%,变异者总生存期更短.NSD 家族表达与多种免疫评分指标显著相关,其中与 IFNG、CD274、免疫排斥评分呈正相关,与免疫评分、ESTIMATE 综合评分呈负相关,并调控 DNA 修复、G2M 检查点、炎症反应、P53 通路等多条信号通路,其中与活性氧上调基因通路和 P53 通路呈负相关.结论 NSD家族在乳腺癌中高表达且存在基因变异,通过调控肿瘤信号通路和免疫微环境影响患者预后,有望成为乳腺癌潜在的生物标志物及治疗靶点.

Objective To investigate the expression patterns,functional roles,and prognostic value of nuclear receptor-binding SET domain(NSD)family members(NSD1,NSD2,NSD3)in breast cancer.Methods The expression levels and prognostic significance of the NSD family in breast cancer were analyzed using the Sangerbox,GEPIA 2.0,HPA,and Kap-lan-Meier Plotter databases.Genetic alterations and survival outcomes were assessed via cBioPortal,and correlations with immune scores were evaluated using TIMER3,and protein-protein interaction networks and associations with cancer-related signaling pathways were examined using the STRING database and TCGA dataset.Results The mRNA and protein expres-sions of NSD1,NSD2,and NSD3 were significantly upregulated in breast cancer,there was correlated expression among fam-ily members.High NSD1 expression was associated with shortened distant metastasis-free survival(DMFS).High NSD2 ex-pression was significantly associated with shortened relapse-free survival(RFS),overall survival(OS),and DMFS in breast cancer patients.High NSD3 expression was slightly associated with longer RFS,but with shorter OS,DMFS and worse prog-nosis.Genetic alteration rates in NSD genes ranged from 1.3%to 12.4%,and alterations were associated with shorter OS.NSD expression showed significant correlations with multiple immune score indicators,including positive correlations with IF-NG,CD274,and exclusion score,and negative correlations with immune score and ESTIMATE score.The NSD family regu-lated various signaling pathways including DNA repair,G2M checkpoint,inflammatory response,and P53 pathway,among which it was negatively correlated with reactive oxygen species-upregulated gene pathway and P53 pathway.Conclusion NSD family is highly expressed in breast cancer and exhibits genetic variations.It influences patient prognosis by regulating tumor signaling pathways and the immune microenvironment,and is expected to become a potential biomarker and therapeutic target for breast cancer.
