新疆南美白对虾副溶血性弧菌的分离鉴定OA

为了解新疆主要养殖区南美白对虾副溶血性弧菌的感染情况,2025年6-9月从新疆巴州、昌吉、塔城等主要产区12家养殖场的47口疑似发病池塘中,采集187份南美白对虾样品(包括虾苗、幼虾和成虾),采用TCBS琼脂培养基进行弧菌分离纯化,通过16S rDNA序列分析进行菌种鉴定;同时,为分析不同样品处理方式对检测结果的影响,分别对肝胰腺、肝胰腺增菌液和分离菌株提取DNA进行PCR检测,对检测结果进行比较分析.结果显示:副溶血性弧菌总检出率为21.4%(40/187),共分离出73株副溶血性弧菌;分离菌株与副溶血性弧菌标准菌株的序列同源性高达99%以上,证实了鉴定结果的准确性;经对不同样品进行检测,分离菌株PCR检出率(21.4%)最高,肝胰腺增菌液PCR检出率(19.8%)高于肝胰腺(9.6%).结果表明:新疆地区南美白对虾中存在副溶血性弧菌感染;对于低载菌量样品检测和快速筛查场景,建议采用提取肝胰腺增菌液DNA进行PCR检测.

In order to identify infection status of Vibrio parahaemolyticus(V.parahaemolyticus)in Litopenaeus vannamei(L.vannamei)in major farming areas of Xinjiang,187 shrimp samples(including post-larvae,juveniles and adults)were collected from 47 ponds where disease outbreaks were suspected on 12 farms in major production areas of Bayingolin,Changji and Tacheng from June to September 2025,to isolate and purify vibrio species using TCBS agar medium,and to identify bacterial strains through 16S rDNA sequence analysis;concurrently,DNAwas extracted from hepatopancreas,hepatopancreas enrichment broth and the isolated strain for PCR assay to compare and analyze the test results.The results showed that the overall detection rate of V.parahaemolyticus was 21.4%(40/187),with a total of 73 strains isolated;over 99%sequence homology was observed between the isolated strain and the standard strain,attesting that the identification was accurate;based on the results of different samples,the detection rate by PCR was the highest for isolated strain(21.4%),followed by hepatopancreas enrichment broth(19.8%),and then hepatopancreas(9.6%).In conclusion,V.parahaemolyticus infection was available in L.vannamei in Xinjiang;and DNA extracted from hepatopancreas enrichment broth was recommended for PCR assay for samples with low bacterial loads and in rapid screening scenarios.
