新型鹅细小病毒VP3病毒样颗粒的制备及免疫原性鉴定OA
Preparation and Immunogenicity Identification of VP3 Virus-like Particles of Novel Goose Parvovirus
新型鹅细小病毒(novel goose parvovirus,NGPV)是一种可引起鸭短喙与侏儒综合征的无包膜单链DNA病毒.为制备NGPVVP3病毒样颗粒(VLPs)并鉴定其免疫原性,以PCR扩增NGPV SD株基因组VP3基因,随后对该基因序列进行优化(密码子偏好性优化,同时在目的基因5'端添加翻译增强序列),采用分子伴侣共表达策略构建表达VP3蛋白的重组菌;对表达的VP3蛋白进行纯化,经乳化后制备亚单位疫苗;将制备的疫苗样品免疫雏鸭,评价其免疫效力.结果显示:经透射电镜分析发现,纯化后的VP3蛋白能自组装成直径约20nm、形态均一的VLPs;疫苗免疫效力评价发现,该VLPs与NGPV阳性血清具有高亲和力(抗原效价1:512),且制备的亚单位疫苗能诱导靶动物产生高水平的中和抗体[效价(6.20±0.65)log2].结果表明,本研究成功建立了一套高效的NGPV VP3蛋白可溶性表达及VLPs制备工艺,为NGPV亚单位疫苗的进一步开发与应用奠定了基础.
Short beak and dwarfism syndrome(SBDS)in ducks could be caused by novel goose parvovirus(NGPV)that is a non-enveloped and single-stranded DNA virus.In order to prepare NGPV VP3 virus-like particles(VLPs)and evaluate their immunogenicity,VP3 gene of NGPV SD strain genome was amplified by PCR.The gene sequence was subsequently optimized(through codon preference optimization with addition of a translation-enhancing sequence at 5'end of the target gene),and a recombinant strain expressing the VP3 protein was constructed using a molecular chaperone co-expression strategy.The expressed VP3 protein was purified and emulsified for preparation of subunit vaccines.The prepared vaccine samples were inoculated into ducklings for assessment of their immune efficacy.The results showed that uniform VLPs approximately 20 nm in diameter were assembled from the purified VP3 protein,as confirmed by transmission electron microscopy.Immunogenicity analysis revealed that these VLPs exhibited high affinity with NGPV-positive serum(antigen titer 1∶512),and the prepared subunit vaccines induced high levels of neutralizing antibodies in target animals[titer:(6.20±0.65)log2].In conclusion,an efficient process for soluble expression of NGPV VP3 protein and preparation of VLPs was established,laying a foundation for further development and application of NGPV subunit vaccines.
