[目的]系统比较可育系'宁杞1号'和不育系'宁杞5号'花药形态与小孢子发育的对应关系,明确枸杞小孢子败育的关键时期,并揭示两者超微结构的差异特征,为阐明宁夏枸杞雄性不育的分子调控机制提供理论依据.[方法]以宁夏枸杞可育系'宁杞1号'和不育系'宁杞5号'不同发育阶段的花蕾为试材,对小孢子发育过程进行表型观测和对比分析,并采用压片法和透射电镜技术对花粉母细胞减数分裂过程及小孢子败育结构特征进行观察.[结果]两种枸杞花粉母细胞减数分裂过程中染色体行为正常,均可形成正常的四分体.'宁杞1号'形成的四分体继续发育,最后形成充满淀粉粒和脂滴的可育花粉粒;'宁杞5号'四分小孢子在胼胝质中逐渐收缩、变形、解体,且超微结构显示'宁杞5号'小孢子在胼胝质中形成初生花粉外壁,但靠近细胞壁的细胞质逐渐液泡化,细胞核凝集,最终完全解体消失.另外'宁杞5号'绒毡层细胞的细胞壁还出现异常加厚现象,这也是其细胞程序性死亡异常的一个重要特征.[结论]不育花药败育时期发生在四分体时期,小孢子外的胼胝质不降解是败育的直接原因.推测由于绒毡层细胞发育异常,胼胝质酶缺乏或酶活性不足导致四分小孢子不能游离,最终引起雄性不育.

[Objective]The study aims to systematically compare the correlation between anther morphol-ogy and microspore development in the fertile line'Ningqi 1'and the male sterile line'Ningqi 5',identify the critical stage of microspore abortion,and reveal the differences in ultrastructure between the two lines,thereby providing a theoretical basis for clarifying the molecular regulatory mechanism of male sterility in L.barbarum L..[Method]Flower buds at different developmental stages of the fertile line'Ningqi 1'and the sterile line'Ningqi 5'were used as experimental materials.Phenotypic observations and comparative analyses of micro-spore development were performed.The meiotic process of pollen mother cells and the structural characteristics of microspore abortion were analyzed using the squash methods and transmission electron microscopy.[Result]During the meiosis in pollen mother cells of both lines,chromosome behavior was normal,yielding normal tet-rads.Tetrads in'Ningqi 1'developed into fertile pollen grains filled with starch granules and lipid droplets.In contrast,microspores in'Ningqi 5'underwent progressive shrinkage,deformation,and disintegration within the callose.Ultrastructural analysis revealed that microspores in'Ningqi 5'could form a primary pollen exine within the callose,but the cytoplasm adjacent to the cell wall gradually vacuolated,the nuclei condensed,and eventually the microspores completely disintegrated,leading to pollen abortion.Additionally,the cell walls of the tapetum cells in'Ningqi 5'exhibited abnormal thickening,which indicated abnormal programmed cell death.[Conclu-sion]Anther abortion in sterile lines occurs at the tetrad stage,and the failure of callose degradation around mi-crospores is the direct cause of abortion.It is speculated that abnormal development of tapetum cells results in callase deficiency or insufficient enzyme activity,thereby preventing the release of tetrad microspores and ulti-mately causing male sterility.
