Comprehensive evaluation of low nitrogen tolerance in 7 poplar clones

[目的]探究低氮胁迫对7个白杨无性系幼苗生长和生理的影响,并从中筛选出耐低氮性较强的优良白杨无性系.[方法]采用盆栽试验,以84K杨、毛白杨30号、秦白杨1号、秦白杨3号、秦白杨5号、西北杨1号和西北杨3号7个白杨无性系幼苗为材料,用1 mmol/L NH4NO3(对照组,CK)和0.01 mmol/L NH4NO3(低氮,LN)进行氮处理,在处理60 d后测定各白杨无性系幼苗的生长及生理指标,并通过相关性分析、主成分分析、隶属函数分析对7个白杨组无性系幼苗的耐低氮性进行综合评价.[结果]与CK相比,低氮处理7个白杨无性系幼苗的生长指标(苗高增长量、地径增长量、植株生物量)和叶绿素含量、净光合速率、氮积累(NH4+-N、NO3--N、NO2--N)以及多种酶(硝酸还原酶(NR)、亚硝酸还原酶(NiR)、谷氨酸合成酶(GOGAT)、谷氨酰胺合成酶(GS))活性均明显下降.低氮胁迫下,7个白杨无性系幼苗的叶片和根系中NH4+-N含量下降幅度明显高于NO3--N、NO2--N,表明7个无性系为更偏向于喜铵的树种.相关性分析结果显示,白杨无性系幼苗的生长和生理指标之间有明显相关性.主成分分析将所有测定指标综合成3个成分因子,并将其分为氮积累因子和氮代谢因子.隶属函数分析结果显示,供试的7个白杨无性系幼苗耐低氮性由强到弱的顺序是84K杨>西北杨3号>西北杨1号>秦白杨1号>秦白杨5号>秦白杨3号>毛白杨30号.[结论]7个白杨无性系幼苗中,84K、西北杨3号和西北杨1号的耐低氮性较强,可作为土壤贫瘠地区的主要推广和人工造林树种.

[Objective]This research aimed to explore the growth and physiological response of 7 poplar clones under low nitrogen stress,and to select excellent poplar clones with strong low nitrogen tolerance from them.[Method]A pot experiment was conducted on 7 poplar clones,including 84K,Maobaiyang 30,Qinbai-yang 1,Qinbaiyang 3,Qinbaiyang 5,and Xibeiyang 1 and Xibeiyang 3.Seedlings were subjected to two nitrogen treatments:1 mmol/L NH₄NO₃(control,CK)and 0.01 mmol/L NH₄NO₃(low nitrogen,LN).After 60 days of treatment,growth and physiological parameters of poplar clones seedling were measured.Comprehensive evaluation of low nitrogen tolerance was conducted using correlation analysis,principal component analysis,and membership function analysis.[Result]Compared with CK,the growth indicators(seedling height increment,ground diameter increment,plant biomass),chlorophyll content,net photosynthetic rate,nitrogen accumulation(NH4+-N,NO3--N,NO2--N),and the activities of various enzymes(nitrate reductase(NR),nitrite reductase(NiR),glutamate synthase(GOGAT),and glutamine synthase(GS))of 7 poplar clones under low nitrogen treatment were significantly reduced.Under low nitrogen stress,the decrease in NH4+-N content in the leaves and roots of 7 poplar clones seedlings was significantly higher than that of NO3--N,NO2--N,indicating that the 7 clones were more inclined towards ammonium loving tree species.The correlation analysis results showed that there was a significant correlation between the growth and physiological indicators of poplar clonal seedlings.Principal component analysis integrated all measurement indicators into three component factors and divided them into nitrogen accumulation factors and nitrogen metabolism factors.The results of the membership function analysis showed that the order of low nitrogen tolerance of the 7 tested poplar clones from strong to weak was 84K>Xibeiyang 3>Xibeiyang 1>Qinbaiyang 1>Qinbaiyang 5>Qinbaiyang 3>Maobaiyang 30.[Conclu-sion]Among the 7 clones of poplar seedlings,84K,Xibeiyang 3,and Xibeiyang 1 have strong low nitrogen toler-ance and can be used as the main promotion and artificial afforestation tree species in soil poor areas.
