miR-20a-5p在民猪前脂肪细胞分化和产热中的作用OA
Study on the role of miR-20a-5p in preadipocyte differentiation and thermogenesis in Min pigs
[目的]以民猪为研究对象,对 miR-20a-5p在民猪各组织器官和不同部位脂肪中的表达及其对体外培养的前脂肪细胞分化和产热相关基因的影响进行了研究,以揭示其在民猪抗寒性状形成中的作用.[方法]用 RT-qPCR法检测 miR-20a-5p在民猪心脏、肝脏、脾脏、肺脏、肾脏、背部肌肉和背部脂肪等7个组织器官以及不同温度环境下背部、腹部、颈部、臀部、胸部和腹股沟等6个不同部位脂肪组织中的表达水平;同时,在体外培养的民猪前脂肪细胞中过表达 miR-20a-5p,检测其对前脂肪细胞分化和重要产热通道中关键基因表达的影响.[结果]miR-20a-5p在民猪肝脏和背肌中的表达量极显著高于其他器官,而在胸部脂肪中的表达量则极显著高于其他部位脂肪;民猪经寒冷刺激后,miR-20a-5p在背部和颈部脂肪中的表达量极显著升高,而在腹股沟脂肪中的表达量极显著降低;在体外培养的民猪前脂肪细胞中过表达 miR-20a-5p会极显著促进前脂肪细胞中脂滴形成和米色化分化相关基因表达,显著抑制 UCP3途径产热相关基因的表达,提高Ca2+循环途径和肌酸循环途径关键基因的表达.[结论]miR-20a-5p在猪的各组织器官和不同部位脂肪中普遍表达,但表达量存在明显差异;在寒冷刺激下,不同部位脂肪组织中 miR-20a-5p的表达量不同;miR-20a-5p能够促进民猪前脂肪细胞米色化,抑制 UCP3途径产热,促进Ca2+循环途径和肌酸循环途径产热.
[Objective]This study investigated the expression of miR-20a-5p in various tissues and fat deposits in different parts of the Min pig,as well as its influence on the differentiation of pre-adipocytes and thermogenesis-related genes,to elucidate its role in the development of cold resistance traits in Min pigs.[Method]The expression levels of miR-20a-5p were compared across seven tissues and organs(the heart,liver,spleen,lungs,kidneys,back muscle,and back fat)and the fat tissues from six distinct body parts(back,abdomen,neck,buttocks,chest,and inguinal area)under varying temperature conditions,with RT-qPCR as the tool.Furthermore,miR-20a-5p was overexpressed in pre-adipocytes cultured in vitro from Min pigs to assess its impact on pre-adipocyte differentiation and key gene expression in a significant ther-mogenic pathway.[Result]The expression of miR-20a-5p was significantly higher in the liver and back muscles compared to other organs,while its expression in chest fat was extremely significantly higher than that in fat from other parts.After Min pigs were subjected to cold stimulation,there was a significant in-crease in the expression of miR-20a-5p in the fat of the back and neck of Min pigs,and a significant de-crease in the inguinal fat.Overexpression of miR-20a-5p in pre-adipocytes cultured in vitro significantly en-hanced lipid droplet formation and beige adipocyte differentiation-related gene expression,significantly in-hibited the UCP3 pathway thermogenesis-related gene expression,and increased the expression of key genes of the Ca2+cycling pathway and the creatine futile cycle pathway.[Conclusion]miR-20a-5p is ubi-quitously expressed across various tissues and organs in pigs.However,its expression levels demonstrate significant difference.When pigs are subjected to cold stimulation,the miR-20a-5p expression in fat tissues from different parts exhibits difference in their expression levels.In Min pigs,miR-20a-5p can induce the beige of preadipocytes,impede heat production via the UCP3 pathway,and concurrently,enhance heat pro-duction through both the Ca2+cycling and creatine futile cycle pathways.
