目的 探讨川陈皮素调节 Hippo/YAP 信号通路对高糖诱导的人视网膜血管内皮细胞损伤的影响.方法 将人视网膜微血管内皮细胞(HRMEC)分为对照组、高糖组、L-川陈皮素组、M-川陈皮素组、H-川陈皮素组、川陈皮素+PY-60 组.对照组HRMEC 在浓度为5.5 mmol·L-1葡萄糖条件下培养,同时补充25.0 mmol·L-1甘露醇以达到相同的渗透压,高糖组、L-川陈皮素组、M-川陈皮素组、H-川陈皮素组、川陈皮素+PY-60 组HRMEC 在含25.0 mmol·L-1葡萄糖的培养基中培养.48 h后,对照组、高糖组HRMEC采用正常培养基培养,L-川陈皮素组、M-川陈皮素组、H-川陈皮素组分别采用含16 μmol·L-1、32 μmol·L-1、64 μmol·L-1 川陈皮素的培养基培养,川陈皮素+PY-60组采用含64 μmol·L-1川陈皮素和 10 μmol·L-1 PY-60 的培养基培养.CCK-8 测定各组细胞的相对细胞活力;基质胶观察各组细胞的血管生成;Transwell 实验检测各组细胞的迁移能力;二氢罗丹明 123 测定各组细胞的活性氧(ROS)水平;JC-1 染色测定各组细胞的线粒体膜电位变化;Western blot 法检测各组细胞 P-YAP1 蛋白表达.结果 与对照组相比,高糖组 HRMEC 的相对细胞活力、JC-1 比值、p-YAP1 蛋白水平均降低,血管形成分支数和迁移细胞数增加,ROS 相对荧光强度提高(均为 P<0.05);与高糖组相比,L-川陈皮素组、M-川陈皮素组、H-川陈皮素组 HRMEC 的相对细胞活力、JC-1比值、p-YAP1 蛋白水平逐渐提高,血管形成分支数和迁移细胞数逐渐减少,ROS 相对荧光强度逐渐降低(均为 P<0.05);与 H-川陈皮素组相比,川陈皮素+PY-60 组HRMEC 的相对细胞活力降低,血管形成分支数增加,迁移细胞数增加,ROS 相对荧光强度提高,JC-1 比值降低,p-YAP1 蛋白表达降低(均为 P<0.05).结论 川陈皮素通过激活 Hippo 途径,促进 YAP1 磷酸化减轻高糖诱导的 HRMEC 损伤.

Objective To investigate the effect of nobiletin on high glucose-induced injury of human retinal vascular endothelial cells by adjusting the Hippo/YAP signaling pathway.Methods Human retinal microvascular endothelial cells(HRMEC)were divided into the control group,the high glucose group,the L-nobiletin group,the M-nobiletin group,the H-nobiletin group,and the nobiletin+PY-60 group.HRMECs in the control group were cultured under a glucose concentration of 5.5 mmol·L-1,with 25.0 mmol·L-1 mannitol added to achieve equivalent osmotic pressure.HRMECs in the high glu-cose group,L-nobiletin group,M-nobiletin group,H-nobiletin group,and nobiletin+PY-60 group were cultured in medium con-taining 25.0 mmol·L-1 glucose.After 48 hours,HRMECs in the control and high glucose groups were cultured in normal medium;those in the L-nobiletin,M-nobiletin,and H-nobiletin groups were cultured in medium containing 16 μmol·L-1,32 μmol·L-1,and 64 μmol·L-1 nobiletin,respectively;and those in the nobiletin+PY-60 group were cultured in medium containing 64 μmol·L-1 nobiletin and10 μmol·L-1 PY-60.Cell viability in each group was assessed using the CCK-8 assay;angiogen-esis was observed using Matrigel;cell migration was evaluated using the Transwell assay;reactive oxygen species(ROS)levels were measured using active dihydrorhodamine 123;changes in mitochondrial membrane potential were determined using JC-1 staining;and p-YAP1 protein expression was detected by Western blot.Results Compared with the control group,the high glucose group exhibited decreased HRMEC viability,JC-1 ratio,and p-YAP1 protein level,increased number of angiogenic branches and migrating cells,and elevated ROS fluorescence intensity(all P<0.05).Compared with the high glucose group,the L-nobiletin,M-nobiletin,and H-nobiletin groups showed gradually increased HRMEC viability,JC-1 ratio,and p-YAP1 protein level,gradually decreased number of angiogenic branches and migrating cells,and gradually reduced ROS fluorescence intensity(all P<0.05).Compared with the H-nobiletin group,the nobiletin+PY-60 group showed de-creased relative cell viability of HRMECs,increased number of angiogenic branches,increased number of migrating cells,in-creased relative ROS fluorescence intensity,decreased JC-1 ratio,and decreased level of p-YAP1 protein expression(all P<0.05).Conclusion Nobiletin alleviates high glucose-induced HRMEC injury by activating the Hippo pathway and promo-ting YAP1 phosphorylation.
