薯蓣皂苷激活Nrf2/HO-1通路在糖尿病视网膜病变大鼠发病机制中的作用OA
Effect of dioscin on the pathogenesis of diabetic retinopathy in rats by activa-ting the nuclear factor E2-related factor 2/heme oxygenase-1 pathway
目的 探讨薯蓣皂苷(Dio)激活核因子 E2 相关因子 2(Nrf2)/血红素加氧酶-1(HO-1)通路在糖尿病视网膜病变(DR)大鼠发病机制中的作用.方法 将大鼠分为对照(NC)组、DR 组、Dio 低剂量组(Dio-L)组、Dio 高剂量(Dio-H)组、Dio-H+Nrf2 抑制剂(ML385)组,除 NC 组外其余各组均通过腹腔注射链脲佐菌素(STZ)诱导 DR 大鼠模型.测定空腹状态下大鼠血糖水平和体重;HE 染色观察大鼠视网膜组织病理变化情况;TUNEL 染色检测大鼠视网膜组织细胞凋亡情况;试剂盒检测大鼠视网膜组织中谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)水平;qRT-PCR 实验检测大鼠视网膜组织中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6、NAD(P)H:醌氧化还原酶1(NQO1)、谷氨酸-半胱氨酸连接酶催化亚基(GCLC)水平;Western blot 检测大鼠视网膜组织中 Nrf2、HO-1 蛋白水平.结果 NC 组大鼠视网膜形态正常,细胞排列整齐且无炎性浸润;与 NC 组相比,DR 组大鼠视网膜神经节细胞数量减少,炎性浸润伴水肿空泡化;空腹血糖、细胞凋亡率、MDA,TNF-α、IL-1β、IL-6 mRNA 表达水平均明显升高(均为 P<0.05),体重、GSH-Px、CAT 水平,NQO1、GCLCmRNA 表达水平,Nrf2、HO-1 蛋白表达水平均明显降低(均为 P<0.05).与 DR 组相比,Dio-L 组、Dio-H 组大鼠视网膜神经节细胞数量均明显增加,炎性浸润程度均减轻;空腹血糖、细胞凋亡率、MDA,TNF-α、IL-1β、IL-6 mRNA 表达水平均明显降低(均为 P<0.05),体重、GSH-Px、CAT 水平,NQO1、GCLCmRNA 表达水平,Nrf2、HO-1 蛋白表达水平均明显升高(均为 P<0.05),且 Dio-H组各指标变化趋势更明显.而在 Dio-H+ML385 组中以上各指标变化趋势均与 Dio-H 组的变化趋势相反(均为 P<0.05).结论 Dio 可通过激活 Nrf2/HO-1 通路抑制氧化应激和炎症反应从而缓解大鼠 DR.
Objective To explore the effect of dioscin(Dio)on the pathogenesis of diabetic retinopathy(DR)in rats by activating the nuclear factor E2-related factor 2(Nrf2)/heme oxygenase-1(HO-1)pathway.Methods The rats were divided into the control(NC)group,the DR group,the low-dose Dio(Dio-L)group,the high-dose Dio(Dio-H)group,and the Dio-H+Nrf2 inhibitor(ML385)group.Except for the NC group,all other groups were induced to establish a DR rat model via intraperitoneal injections of streptozotocin.Fasting blood glucose levels and body weight of rats were measured.Hematoxylin-eosin staining was used to observe pathological changes in retinal tissue of rats;TUNEL staining was employed to detect cell apoptosis in retinal tissue of rats;assay kits were applied to measure the levels of glutathione perox-idase(GSH-Px),catalase(CAT),and malondialdehyde(MDA)in retinal tissue of rats;quantitative real-time polymerase chain reaction was performed to assess the levels of tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,NAD(P)H:quinone oxidoreductase 1(NQO1),and glutamate-cysteine ligase catalytic subunit(GCLC)in retinal tissue of rats;and Western blot was used to determine protein levels of Nrf2 and HO-1 in retinal tissue of rats.Results In the NC group,rat retina exhibited normal morphology,with well-arranged cells and no inflammatory infiltration.Compared to the NC group,the DR group showed a significant reduction in retinal ganglion cells,accompanied by inflammatory infiltration and vacuolar edema;in addition,fasting blood glucose level,apoptosis rate,MDA,TNF-α,IL-1β,and IL-6 mRNA levels were significantly elevated(all P<0.05),while body weight,GSH-Px and CAT levels,NQO1 and GCLC mRNA levels,as well as Nrf2 and HO-1 protein levels markedly decreased(all P<0.05).In contrast to the DR group,both the Dio-L and Dio-H groups showed a significant increase in retinal ganglion cells and reduced inflammatory infiltration;furthermore,fasting blood glucose level,apoptosis rate,MDA,TNF-α,IL-1β,and IL-6 mRNA levels significantly decreased(all P<0.05),whereas body weight,GSH-Px and CAT levels,NQO1 and GCLC mRNA levels,as well as Nrf2 and HO-1 protein levels were markedly elevated(all P<0.05),with more pronounced changes in the Dio-H group.In the Dio-H+ML385 group,the trend of changes in all the above indicators was opposite to that in the Dio-H group(all P<0.05).Conclusion Dio can alleviate DR in rats by activating the Nrf2/HO-1 pathway to inhibit oxidative stress and inflammatory response.
