目的 探究 Bcl2 样蛋白 12(Bcl2L12)对实验性自身免疫性葡萄膜炎(EAU)小鼠光感受器细胞间维生素 A 类结合蛋白 1-20(IRBP1-20)特异性辅助性 T 细胞17(Th17)的影响.方法 将12 只8~10 周龄 C57BL/6J 雌鼠分为 EAU 组和正常对照组,每组6 只.EAU 组小鼠腹腔注射百日咳毒素后,用 IRBP1-20 与完全弗氏佐剂乳化液注射足垫及背部皮下建立模型,对照组不接受免疫处理.免疫后 13 d 评估模型构建情况.分离 EAU 小鼠脾脏及淋巴结中的 T 细胞,实时荧光定量 PCR 检测Bcl2L12 mRNA 表达水平.利用小鼠 T 细胞淋巴瘤细胞系(EL4)细胞筛选靶向Bcl2L12 的有效小干扰 RNA(siRNA),将筛选出的 siBcl2L12 或阴性对照(si-NC)转染至 EAU 小鼠 CD4+T 细胞,与经 γ 射线照射后的抗原呈递细胞共培养,加入白细胞介素(IL)-23 诱导 Th17 极化.实时荧光定量 PCR 检测 Th17 相关基因[IL-17A、维甲酸相关孤儿受体 γt(RORγt)、IL-1 受体(IL-1R)、IL-23 受体(IL-23R)及粒细胞-巨噬细胞集落刺激因子(GM-CSF)等]的 mRNA 表达量,ELISA 检测细胞上清中 IL-17 水平,流式细胞仪检测 Th17 细胞比例.结果 免疫后 13 d,EAU 组小鼠眼底出现炎症改变,模型构建成功.与对照组相比,EAU 组小鼠 CD4+T 细胞中 Bcl2L12 mRNA 表达显著上调(P<0.01).EL4 细 胞 验 证 结 果 显 示,siBcl2L12-2 敲低效率最高(P<0.01),用于后续实验.在共培养体系中,与 Blank 组和 si-NC 组相比,siBcl2L12 组Th17 细胞比例显著降低(P<0.05),上清中 IL-17 浓度显著下降(P<0.05),且 IL-17A、RORγt、IL-1R、IL-23R、GM-CSF mRNA 相对表达量均明显降低(均为 P<0.01).结论 敲低 T 细胞中的 Bcl2L12 可以抑制 EAU 小鼠 IRBP1-20 特异性 Th17 细胞的极化及相关炎症因子的表达,Bcl2L12 对 IRBP1-20特异性 Th17 细胞反应具有正向调控作用.

Objective To investigate the effect of Bcl2-like protein 12(Bcl2L12)on interphotoreceptor retinoid-binding protein 1-20(IRBP1-20)-specific T helper 17(Th17)cells in mice with experimental autoimmune uveitis(EAU).Methods Twelve female C57BL/6J mice(8-10 weeks old)were divided into an EAU group and a control group,with 6 mice in each group.Mice in the EAU group received an intraperitoneal injection of pertussis toxin,followed by subcutaneous injections of IRBP1-20 and complete Freund's adjuvant emulsion into the footpads and back to establish the model,while mice in the control group received no immunization.At day 13 post-immunization,model establishment was evaluated.T cells were isolated from the spleen and lymph nodes of EAU mice,and Bcl2L12 mRNA expression level was detected by quantita-tive real-time polymerase chain reaction(qRT-PCR).Effective small interfering RNAs targeting Bcl2L12 were screened out using the mouse T lymphoma cell line(EL4)cells.The selected siBcl2L12 or negative control(si-NC)was transfected into CD4+T cells in EAU mice,which were then co-cultured with γ-irradiated antigen-presenting cells.Interleukin(IL)-23 was added to induce Th17 polarization.The mRNA expression levels of Th17-associated genes[IL-17A,retinoid-related orphan receptor γt(RORγt),IL-1 receptor(IL-1R),IL-23 receptor(IL-23R),and granulocyte-macrophage colony-stimulating factor(GM-CSF)]were detected by qRT-PCR,IL-17 levels in the cell supernatant were measured by ELISA,and the proportion of Th17 cells was analyzed by flow cytometry.Results At day 13 post-immunization,EAU mice exhibited inflammatory changes in the fundus,confirming successful model establishment.Compared with the control group,Bcl2L12 mRNA expres-sion was significantly upregulated in CD4+T cells in EAU mice(P<0.01).EL4 cell validation results showed that siBcl2L12-2 exhibited the highest knockdown efficiency(P<0.01)and was used for subsequent experiments.In the co-cul-ture system,compared with the Blank group and si-NC group,the si-Bcl2L12 group showed a significantly reduced propor-tion of Th17 cells(P<0.05),significantly decreased IL-17 levels in the supernatant(P<0.05),and markedly downregulat-ed mRNA expression levels of IL-17A,ROR-γt,IL-1R,IL-23R,and GM-CSF(all P<0.01).Conclusion Knockdown of Bcl2L12 in T cells inhibits the polarization of IRBP1-20-specific Th17 cells and the expression of related inflammatory factors in EAU mice,suggesting that Bcl2L12 positively regulates IRBP1-20-specific Th17 cell responses.
