目的:探讨银杏去甲基酮酯对恒河猴激光致脉络膜新生血管(choroidal neovascularization,CNV)生成的抑制作用.方法:16 只健康恒河猴进行双眼眼底激光光凝形成 CNV 造模后,设安慰剂组(n=6)、康柏西普眼内注射液组(n=3)、银杏叶提取物组(n=3)、银杏去甲基酮酯组(n=4).分别在造模第 21 天(D0)给药前及给药后的第 28 天(D28)进行眼底摄影(fundus photography,FP)、眼底荧光素血管造影(fundus fluorescein angiography,FFA)、光学相干断层扫描(optical coherence tomography,OCT)检查;主要疗效指标包括给药前后Ⅳ级光斑数量、Ⅳ级光斑荧光渗漏面积变化率,Ⅳ级光斑视网膜厚度变化率.结果:Ⅳ级光斑数量、Ⅳ级光斑荧光渗漏面积变化率和Ⅳ级光斑视网膜厚度变化观测指标,安慰剂组结果分别为 30 个、(101.7±84.3)%、(64.7±91.5)%,康柏西普眼内注射液组结果分别为 20 个、(-100.0±0.0)%、(-94.1±14.3)%,银杏叶提取物组分别为 24 个,(19.9±76.23)%、(3.0±102.03)%,银杏去甲基酮酯组结果分别为 26 个、-(34.7±53.3)%、-(58.6±48.5)%,银杏去甲基酮酯组与安慰剂组比较差异均有统计学意义(P<0.05).结论:银杏去甲基酮酯对恒河猴 CNV 模型的荧光渗漏和视网膜增厚有显著的改善作用.

Objective:To explore the inhibitory effect of Ginkgo Demethylketone Ester on laser-induced choroidal neovascularization(CNV)in rhesus monkeys.Methods:Sixteen healthy rhesus monkeys received bilateral fundus laser photocoagulation to establish CNV models.The subjects were divided into four groups:the placebo group(n=6),the conbercept injection group(n=3),the Ginkgo leaf extract group(n=3),and the Ginkgo Demethylketone Ester group(n=4).Fundus photography(FP),fundus fluorescein angiography(FFA),and optical coherence tomography(OCT)were conducted at baseline(D0,before administration)and on day 28(D28)after administration.The primary efficacy endpoints included:1)the number of grade IV lesions,2)the change rate of fluorescein leakage area in grade IV lesions,and 3)the change rate of retinal thickness in grade IV lesions.Results:In Placebo group,there were 30 grade IV lesions.The change rates of the fluorescein leakage area and retinal thickness were(101.7±84.3)%and(64.7±91.5)%respectively.In conbercept group(0.5 mg/eye),there were 20 grade IV lesions.The change rates of fluorescein leakage area and retinal thickness were(-100.0±0.0)%,and(-94.1±14.3)%respectively.In Ginkgo leaf extract group,there were 24 grade IV lesions.The change rates of the fluorescein leakage area and retinal thickness were(-19.9±76.23)%,and(3.0±102.03)%respectively.In Ginkgo Demethylketone Ester group,there were 26 grade IV lesions.The change rates of the fluorescein leakage area and retinal thickness were(-34.7±53.3)%,and(-58.6±48.5)%respectively.The Ginkgo Demethylketone Ester group showed statistically significant differences from the placebo group in all endpoints.Conclusions:Ginkgo Demethylketone Ester significantly improved fluorescein leakage and retinal thickening in the rhesus monkey CNV model.
