目的:探讨磷酸二酯酶3(PDE3)特异性抑制剂米利酮体外对正常人精子与弱精子症患者精子运动能力的影响.方法:采用10 μmol/L米利酮对正常人精子和弱精子症患者精子进行体外处理1 h,未经米利酮处理的正常人精子和弱精子症患者精子作为对照组,通过计算机辅助精子分析(CASA)系统评估精子运动参数.结果:与未经米利酮处理的正常人精子比较,米利酮处理后的正常人精子的总运动、前向运动、非前向运动以及运动参数均无影响(P>0.05);与未经米利酮处理的弱精子症患者精子比较,米利酮处理后可显著提升精子的总运动、前向运动和非前向运动,并增强精子平均曲线运动速度、平均直线运动速度、平均路径运动速度及平均鞭打频率(P<0.05),2组精子头部侧摆度及运动模式(平均侧摆幅值、平均移动角度、运动的直线性、运动的摆动性、运动的前向性)比较差异无统计学意义(P>0.05).结论:米利酮在体外能有效激活弱精子症患者精子的运动功能.

Objective:To investigate the effects of phosphodiesterase 3(PDE3)specific inhibitor milrinone on the motility of sperm from healthy individuals and asthenozoospermic patients in vitro.Methods:Sperm from healthy individuals and asthenozoospermic patients were treated in vitro with 10 μmol/L milrinone for 1 h.The two types of sperm without milrinone treatment served as the control groups.Sperm motility parameters were assessed using a computer-assisted sperm analysis(CASA)system.Results:Compared with the control,milrinone had no effect on total motility,progressive motility,non-progressive motility and motility parameters in healthy sperm(P>0.05).Compared with the control in asthenozoospermic samples,milrinone significantly increased the total motility,progressive motility,non-progressive motility,and also enhanced the mean curvilinear velocity,mean straight-line velocity,average path velocity,and mean beat-cross frequency of sperm(P<0.05).There were no statistically significant differences between the two groups in terms of sperm lateral head displacement and movement patterns(mean amplitude of lateral head displacement,mean angular displacement,linearity,wobble and straightness)(P>0.05).Conclusion:Milrinone can effectively activate sperm motility in asthenozoospermic patients in vitro.
