超声辅助提取西藏赤芝多糖工艺优化及其降糖活性研究OA

西藏自治区林芝地区独特的高原生态环境孕育了品质良好的灵芝资源.林芝灵芝作为国家地理标志产物,其富含的多糖、三萜等活性物质赋予其独特的保健功效.多糖作为灵芝的主要活性物质之一,使用传统的热水萃取存在效率低、高耗能等问题,而物理辅助技术如超声辅助提取具备绿色高效、条件温和的特点,在活性成分提取领域展现出显著优势.研究以西藏自治区林芝地区人工栽培的赤芝作为实验原料,采用超声辅助提取技术,以赤芝多糖提取率设定为衡量指标,通过单因素实验与正交试验优化提取条件,并探究所得多糖对α-葡萄糖苷酶与α-淀粉酶活性的抑制率.超声辅助提取赤芝多糖的最优条件为:料液比1∶10、超声功率200 W、提取温度70 ℃以及提取时间40 min,在此条件下,多糖提取率可达2.38%.此外,赤芝多糖粗提物对α-葡萄糖苷酶与α-淀粉酶活性的抑制率呈现出明显的浓度依赖性,在粗多糖浓度为1.0 mg/mL时,对α-葡萄糖苷酶与α-淀粉酶活性抑制率分别为72.9%和80.8%,这充分表明赤芝多糖具备良好的降糖活性.本研究不仅为西藏赤芝资源的高值化利用提供关键技术支撑,更为天然降糖药物研发开辟新路径.

In Nyingchi region of Tibet autonomous region,the distinctive plateau ecological en-vironment has nurtured high-quality Ganoderma lucidum resources.As a product with na-tional geographical indication,Ganoderma lucidum from Nyingchi is rich in active substances such as polysaccharides and triterpenoids,which endow it with unique health-care effects.Polysaccharides,being one of the major active substances in Ganoderma lucidum,are faced with issues such as low efficiency and high energy consumption when extracted by the tradi-tional hot water extraction method.In contrast,physical-assisted techniques,like ultrasonic-assisted extraction,feature environmental friendliness,high efficiency,and mild reaction con-ditions,thus demonstrating remarkable advantages in the field of active ingredient extraction.This research utilized artificially cultivated Ganoderma lucidum in Nyingchi Region of Tibet Autonomous Region as the experimental material and employed ultrasonic-assisted extraction technology.The extraction rate of Ganoderma lucidum polysaccharides was set as the evalua-tion criterion.Single-factor experiments and orthogonal experiments were conducted to opti-mize the extraction conditions.Additionally,the inhibitory rates of the obtained polysaccha-rides on the activities of α-glucosidase and α-amylase were explored.The optimal conditions for ultrasonic-assisted extraction of Ganoderma lucidum polysaccharides were determined as follows:the solid-to-liquid ratio was 1∶10,the ultrasonic power was 200 W,the extraction temperature was 70 ℃,and the extraction time was 40 min.Under these conditions,the ex-traction rate of polysaccharides could reach 2.38%.Furthermore,the inhibitory rates of the crude extract of Ganoderma lucidum polysaccharides on the activities of α-glucosidase and α-amylase exhibited an obvious concentration-dependence.When the concentration of the crude polysaccharides was 1.0 mg/mL,the inhibitory rates on the activities of α-glucosidase and α-amylase were 72.9%and 80.8%respectively.This fully indicates that Ganoderma lucidum polysaccharides possess excellent hypoglycemic activity.This study not only provides crucial technical support for the high-value utilization of Ganoderma lucidum resources in Tibet but also paves a new way for the research and development of natural hypoglycemic drugs.
