[目的]明确不同抗寒桑品种霜前、霜后桑叶的营养与活性物质含量及功能活性差异,筛选适合北方抗寒地区食药用开发的优良桑品种.[方法]以北方5个抗寒桑品种为材料,分别采集霜前、霜后的桑叶样品,采用高效液相色谱串联质谱法测定DNJ活性,用福林酚比色法测定多酚类活性成分,用亚硝酸钠-硝酸铝显色法测定总黄酮含量,通过ABTS、FRAP、DPPH法评价桑叶抗氧化能力,同步开展α-糖苷酶抑制活性检测,对检测结果进行霜前后品种间比较、回归分析、综合评价与聚类分析.[结果]霜前后各品种在1-脱氧野尻霉素(DNJ)、黄酮类物质、抗氧化能力(ABTS、FRAP、DPPH)及α-糖苷酶抑制率等方面均存在极显著差异(P<0.01).霜前,龙桑1号的DNJ、槲皮素和芦丁含量最高;辽育8号在绿原酸、异槲皮苷、FRAP、ABTS方面表现突出;辽鲁11号则在总黄酮、DPPH和α-糖苷酶抑制率上最优.霜后,龙桑1号保持最高DNJ含量;辽育8号中绿原酸、FRAP含量最高,辽鲁11号中总黄酮、ABTS、DPPH含量最高,辽鲁11号和辽育8号均在槲皮素、异槲皮苷、α-糖苷酶抑制率方面表现突出.回归分析表明,在霜前和霜后测定的9个指标中,DPPH是影响总黄酮的核心驱动因素.综合评价结果表明,霜前辽鲁11号表现最佳,辽育16号表现最差,霜后辽鲁11号表现最佳,龙桑1号表现最差.聚类分析表明,霜前后品种均呈现地域聚集特点.[结论]辽鲁11号和辽育8号为北方抗寒地区食药用多元开发的优良桑品种,具备较高的开发利用价值.

[Objective]To clarify the differences in the contents of nutrients,bioactive substances and functional activities in mulberry leaves among different cold-resistant mulberry varieties before and after frost,and to screen excellent mulberry varieties suitable for edible and medicinal development in cold-resistant regions of northern China.[Methods]Five northern cold-resistant mulberry varieties were used as materials,and mulberry leaf samples were collected before and after frost,respectively.1-deoxynojirimycin(DNJ)was determined by high performance liquid chromatography-tandem mass spectrometry,polyphenols by Folin-Ciocalteu colorimetry,and total flavonoids by sodium nitrite-aluminum nitrate colorimetry.The antioxidant capacity was evaluated by ABTS,FRAP and DPPH assays,and α-glucosidase inhibitory activity was detected simultaneously.The results were subjected to comparison between varieties before and after frost,regression analysis,comprehensive evaluation and cluster analysis.[Results]There were extremely significant differences(P<0.01)among the varieties in 1-deoxynojirimycin(DNJ),flavonoids,antioxidant capacities(ABTS,FRAP,DPPH),and α-glucosidase inhibition rate before and after frost.Before frost,Longsang1 had the highest contents of DNJ,quercetin and rutin;Liaoyu8 performed prominently in chlorogenic acid,isoquercitrin,FRAP and ABTS;while Liaolu 11 was optimal in total flavonoids,DPPH and α-glucosidase inhibition rate.After frost,Longsang1 maintained the highest DNJ content;Liaoyu 8 had the highest contents of chlorogenic acid and FRAP;Liaolu11 showed the highest contents of total flavonoids,ABTS and DPPH;both Liaolu11 and Liaoyu8 performed excellently in quercetin,isoquercitrin and α-glucosidase inhibition rate.Regression analysis indicated that among the 9 indicators measured before and after frost,DPPH was the core driving factor affecting total flavonoids.The comprehensive evaluation results showed that Liaolu 11 had the best performance and Liaoyu16 had the worst before frost;after frost,Liaolu11 still ranked first while Longsang11 was the poorest.Cluster analysis revealed that the varieties showed geographical aggregation characteristics both before and after frost.[Conclusion]Liaolu11 and Liaoyu8 are excellent mulberry varieties for the diversified food and medicinal development in cold-resistant areas of northern China,with high development and utilization value.
