目的 探究祛风清热、活血养血法治疗过敏性结膜炎的作用机制.方法 基于中医传承辅助系统和网络药理学,发掘中药治疗过敏性结膜炎新组方,分析预测新复方(防风、荆芥、蝉蜕、川芎、黄芩、当归、生地黄、赤芍、菊花、甘草)干预过敏性结膜炎的靶点和作用机制.结果 以 CNKI、维普等数据库作为查找源,按一定要求进行检索并剔除后,中医传承辅助系统平台共纳入 131 首方剂;出现频数排名靠前的单味药为防风、甘草、荆芥、蝉蜕、川芎等;在药物归类中,解表药、清热药和补虚药使用频次最多;药性分析多集中在寒性、温性药物,药味苦、甘和辛出现频次高.网络药理学分析频数排名前 10 的药物(新复方-祛风止痒一号方)活性成分 125 个、作用靶基因 251 个、过敏性结膜炎的疾病靶基因 1154 个,新复方干预过敏性结膜炎的交集靶基因有 53 个;祛风止痒一号方干预过敏性结膜炎的关键靶基因主要包括 MAPK3、MMP9、MMP2、IL-1B、IKBKB 等;祛风止痒一号方主要通过 MAPK 信号通路、AGE-RAGE 信号通路及钙信号通路等影响过敏性结膜炎对细胞氧化应激的反应、转录因子结合及正向调节细胞迁移等多种生理过程.结论 中医治疗过敏性结膜炎多采用祛风清热、活血养血法.祛风止痒一号方可能通过调节 MAPK3、MMP9、MMP2、IL-1B、IKBKB 等基因表达,调控 MAPK、AGE-RAGE 及钙信号等信号通路,发挥治疗过敏性结膜炎作用.

Objective To explore the mechanism of expelling wind and clearing away heat,promoting blood circulation and nourishing blood in the treatment of allergic conjunctivitis.Methods Based on the traditional Chinese medicine(TCM)inheritance assistant system and network pharmacology,the new prescription of TCM for allergic conjunctivitis was explored,to analyze and predict the targets and mechanism of new compound(Saposhnikoviae Radix,Schizonepetae Herba,Cicadae Periostracum,Chuanxiong Rhizoma,Scutellariae Radix,Angelicae Sinensis Radix,Rehmanniae Radix,Paeoniae Radix Rubra,Chrysanthemi Flos,Glycyrrhizae Radix et Rhizoma)in the intervention of allergic conjunctivitis.Result With CNKI,VIP and other databases as the source of search,according to certain requirements for retrieval and elimination,a total of 131 prescriptions were included in the TCM inheritance auxiliary system platform.The most frequently used drugs were Saposhnikoviae Radix,Glycyrrhizae Radix et Rhizoma,Schizonepetae Herba,Cicadae Periostracum,Chuanxiong Rhizoma,etc.The analysis of drug properties mainly focused on cold and warm drugs,and bitter,sweet and pungent drugs appeared frequently.The top 10 drugs by frequency association in network pharmacology analysis(New Compound-Qufeng Zhiyang Yihao decoction)have 125 active ingredients,251 target genes,and 1,154 disease target genes for allergic conjunctivitis.There were 53 intersection target genes for the intervention of new compound in allergic conjunctivitis.The key target genes of Qufeng Zhiyang Yihao decoction in treating allergic conjunctivitis mainly include MAPK3,MMP9,MMP2,IL-1B,IKBKB and so on.Qufeng Zhiyang Yihao prescription mainly affects the response of allergic conjunctivitis to cellular oxidative stress,transcription factor binding and positively regulates cell migration through MAPK signaling pathway,AGE-RAGE signaling pathway and calcium signaling pathway.Conclusion TCM treats the allergic conjunctivitis to adopt dispelling wind and clearing away heat,promoting blood circulation and nourishing blood method.Qufeng Zhiyang Yihao decoction may play a role in the treatment of allergic conjunctivitis by regulating the expression of MAPK3,MMP9,MMP2,IL-1B,IKBKB and other genes,and regulating MAPK,AGE-RAGE and calcium signaling pathways.
