视黄醇棕榈酸酯醇质体的制备及其表征研究OA
Preparation and characterization of retinyl palmitate phytosomes
通过超高压微射流均质技术制备了视黄醇棕榈酸酯醇质体(RP-E).表征结果显示,制备的醇质体呈规则球形或椭球形,且观察到磷脂双分子层结构,平均粒径39.18 nm(PDI=0.143),Zeta电位-37.95 mV,表明体系具有优异的分散稳定性.HPLC分析证实RP含量达4.99%,超速离心法测定包封率高达99.8%.稳定性研究表明,在不同储存条件(常温、光照、-15℃、45℃、交替及5℃)下,醇质体粒径均保持小于50 nm;除45℃及交替条件外,活性成分含量均大于4.5%(降解率小于10%),表现出良好的稳定性.该工艺不仅能有效保护视黄醇衍生物,还展现出优异的工业化生产潜力,为类视黄醇原料的纳米包裹提供了创新解决方案.
Retinyl palmitate phytosomes(RP-E)were prepared using ultra-high-pressure microfluidization.Characterization showed that the phytosomes were spherical or ellipsoidal,exhibiting a phospholipid bilayer structure.The average particle size was 39.18 nm(PDI=0.143),and the zeta potential was-37.95 mV,indicating excellent dispersion stability.HPLC analysis confirmed the RP content of 4.99%,and ultracentrifugation revealed an encapsulation efficiency of 99.8%.Stability studies demonstrated that particle sizes remained below 50 nm under various storage conditions(room temperature,light exposure,-15 ℃,45℃,alternating temperature,and 5℃),and the active ingredient content exceeded 4.5%under all conditions except 45 ℃ and alternating temperature,corresponding to degradation rates below 10%.This process not only effectively protects retinol derivatives but also shows strong potential for industrial-scale production,providing an innovative strategy for nano-encapsulation of retinoid raw materials.
