探讨原花青素B2(PB2)对多柔比星(DOX)诱导的人皮肤成纤维细胞(HSF)线粒体损伤的修复作用及其抗衰老机制.建立DOX诱导的HSF衰老模型,分别用不同浓度PB2进行干预.采用荧光显微成像技术检测活性氧(ROS)水平及线粒体膜电位(ΔΨm);分别通过试剂盒检测ATP含量、免疫荧光法检测DNA损伤标志物γ-H2AX、衰老相关β-半乳糖苷酶(SA-β-gal)染色评估细胞衰老表型.结果表明,PB2处理能较好地拮抗DOX诱导的损伤.其中,2 μmol/L的PB2综合干预效果最为显著,可显著降低细胞内ROS积累(降幅大于50%);线粒体功能同步恢复,ΔΨm提升8.75倍,ATP生成量增加33.08%,同时减少γ-H2AX表达(降低25.53%)并抑制SA-β-gal活性(降低38.20%).PB2通过清除ROS、修复线粒体功能(ΔΨm稳态与ATP合成)及减轻DNA损伤,显著延缓DOX诱导的皮肤细胞衰老,为PB2基抗衰化妆品开发提供理论依据.

The protective effects of proanthocyanidin B2(PB2)against doxorubicin(DOX)-induced mitochondrial dysfunction and cellular senescence in human skin fibroblasts(HSFs)was investigated.A DOX-induced HSF senescence model was established,and cells were treated with varying concentrations of PB2.Reactive oxygen species(ROS)levels and mitochondrial membrane potential(ΔΨm)were assessed using fluorescence microscopy.ATP content was measured using commercial assay kits,DNA damage was evaluated via γ-H2AX immunofluorescence,and senescence-associated β-galactosidase(SA-β-gal)staining was used to characterize senescent phenotypes.PB2 treatment effectively mitigated DOX-induced cellular damage.Among the concentrations tested,2 μmol/L PB2 showed the most pronounced effect,reducing intracellular ROS accumulation by over 50%,restoring mitochondrial function(ΔΨm increased 8.75-fold),and enhancing ATP production by 33.08%.Moreover,γ-H2AX expression decreased by 25.53%,and SA-β-gal activity was inhibited by 38.20%.These findings indicate that PB2 delays DOX-induced cellular senescence by scavenging ROS,restoring mitochondrial function(ΔΨm homeostasis and ATP synthesis),and alleviating DNA damage,providing a theoretical basis for the development of PB2-based anti-aging cosmetic products.
