目的 探讨谷氨酸离子型受体N-甲基-D-天冬氨酸受体亚单位2D(GRIN2D)在结直肠癌(CRC)组织中的表达及意义.方法 收集2016-2018年在青岛大学附属医院接受手术治疗的266例Ⅰ~Ⅲ期原发性CRC病人的癌组织和癌旁对照组织.采用免疫组化方法检测两种组织样本中GRIN2D的表达.构建GRIN2D过表达和敲除的CRC细胞系,评估GRIN2D对CRC细胞增殖的影响.结果 CRC组织中GRIN2D的表达显著高于癌旁对照组织(P=0.001).GRIN2D高表达与更晚的肿瘤TNM分期、更深的肿瘤浸润、淋巴结转移、脉管癌栓有关(x2=3.99~8.08,P<0.05).GRIN2D高表达病人无病生存期较低表达病人更短(x2=6.01,P=0.042).敲除GRIN2D可以通过BAX/BCL-2途径促进CRC细胞凋亡,抑制CRC细胞增殖(t=5.73~25.84,P<0.05).结论 GRIN2D在CRC组织中高表达,敲除GRIN2D可以抑制CRC细胞增殖.GRIN2D有望成为CRC新的分子治疗靶点.

Objective To investigate the expression and significance of glutamate receptor,ionotropic,N-methyl-D-aspar-tate 2D(GRIN2D)in colorectal cancer(CRC)tissue.Methods Cancerous tissue samples and adjacent control tissue samples were collected from 266 patients with stage Ⅰ—Ⅲ primary CRC who underwent surgical treatment in The Affiliated Hospital of Qingdao University from 2016 to 2018.Immunohistochemistry was used to measure the expression of GRIN2D in the two types of samples.CRC cell lines with GRIN2D overexpression or knockout were constructed to assess the effect of GRIN2D on CRC cell proliferation.Results The expression level of GRIN2D in CRC tissue was significantly higher than that in adjacent control tissue(P=0.001).The high expression of GRIN2D was associated with advanced TNM stage,deeper tumor infiltration,lymph node metastasis,and vascular cancer thrombus(x2=3.99-8.08,P<0.05).The patients with high expression of GRIN2D had a signifi-cantly shorter disease-free survival time than those with low expression(x2=6.01,P=0.042).GRIN2D knockout could promote CRC cell apoptosis and inhibit CRC cell proliferation through the BAX/BCL-2 pathway(t=5.73-25.84,P<0.05).Conclusion GRIN2D is highly expressed in CRC tissue,and GRIN2D knockout can inhibit CRC cell proliferation.GRIN2D is expected to be-come a new molecular therapeutic target for CRC.
