饲料中添加辅酶Q10对花鲈抗高温胁迫能力的影响OA
The Influence of Dietary Coenzyme Q10 on the Resistance of Spotted Seabass(Lateolabrax maculatus)to Heat Stress
为开发提高花鲈(Lateolabrax maculatus)抗高温胁迫能力的功能性饲料,在基础饲料中分别添加0、10、20 mg/kg 的辅酶 Q10,分别记为 Con、C10 和 C20.在水温为33℃的高温胁迫环境中,用这3种饲料分别投喂花鲈 56 d.结果表明,在高温胁迫下,C10 和 C20 都可显著提高花鲈的增重率(P<0.05),并缓解花鲈鳃和肝脏组织结构损伤,减少肝脏脂肪沉积,显著提高花鲈总抗氧化能力(total an-tioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的活性(P<0.05),显著降低肝脏丙二醛(malondialdehyde,MDA)含量(P<0.05),缓解氧化应激.进一步研究发现,C10 和 C20 还能增强花鲈线粒体功能,显著提高柠檬酸合成酶(citrate syn-thase,CS)和琥珀酸脱氢酶(succinate dehydrogenase,SDH)的活性(P<0.05),提高线粒体三羧酸循环速率,并显著上调线粒体自噬和生成相关基因 pink1、mul1、atg5、pgc-1α、pgc-1β、nrf1 的相对表达量(P<0.05),促进线粒体的自我更新与修复,维持正常的线粒体功能.因此,在饲料中添加10 或20 mg/kg辅酶 Q10 可以显著提高花鲈抗高温胁迫的能力.
To develop a functional feed that enhances the high-temperature stress resistance of Lateolabrax maculatus,this study focused on L.maculatus and added 0,10 and 20 mg/kg of coenzyme Q10 to the basic feed,denoted as Con,C10 and C20,respectively.In a high-temperature stress environment with a water tempera-ture of 33℃,these three types of feed were fed to sea bass separately for 56 days.The results demonstrated that adding 10 or 20 mg/kg of coenzyme Q10 to the diet could significantly increase the lower weight gain of spotted seabass under heat stress,and alleviate the morphological damage of gill and liver of spotted seabass,reduce the fatty deposition of liver,significantly increase the activities of total antioxidant capacity(T-AOC),superoxide dismutase(SOD),and catalase(CAT)of spotted seabass,and significantly decrease the malondial-dehyde(MDA)content in liver and alleviate oxidative stress.Further studies revealed that coenzyme Q10 also promoted the mitochondrial function of the spotted seabass,significantly increased the activities of citrate syn-thase(CS)and succinate dehydrogenase(SDH),increased the rate of the mitochondrial tricarboxylic acid cy-cle,and significantly up-regulated the relative expression of the mitochondrial autophagy and generation-related genes pink1,mul1,atg5,pgc-1α,pgc-1β and nrf1,which promoted the mitochondrial self-renewal and repair,and maintain normal mitochondrial function.So adding either 10 or 20 mg/kg of coenzyme Q10 to the diet could enhance the spotted seabass's ability to withstand heat stress and significantly improve its growth perform-ance.
