目的 观察参芪黄颗粒对脓毒症大鼠胃肠功能障碍的改善作用.方法 通过盲肠结扎穿孔法建立脓毒症大鼠模型,将大鼠随机分为假手术组、盲肠结扎穿孔组、参芪黄低、中、高剂量组及地塞米松组.造模结束后连续3 天给药,参芪黄低、中、高剂量组予参芪黄颗粒灌胃,地塞米松组予地塞米松灌胃,其余组予等体积蒸馏水灌胃.酶联免疫吸附试验检测血清白细胞介素-6、肿瘤坏死因子-α、抗炎因子白细胞介素-10 和精氨酸酶-1、肠道损伤标志物二胺氧化酶、D-乳酸;苏木精—伊红染色观察结肠组织病理改变;宏基因组学方法测序肠道微生物组,分析组间门物种水平组成及筛选组间特征微生物.结果 参芪黄颗粒能降低血清中白细胞介素-6、肿瘤坏死因子-α 炎性因子水平(P<0.01,P<0.05),提高抗炎因子白细胞介素-10、精氨酸酶-1 水平(P<0.01,P<0.05);改善结肠病理改变,降低肠损伤标志物二胺氧化酶、D-乳酸水平(P<0.01);调节肠道微生物群落结构,恢复菌群平衡.结论 参芪黄颗粒对脓毒症大鼠具有治疗作用,可调节炎症反应,改善肠损伤,调节肠道菌群.

Objective To investigate the efficacy of Shenqihuang Granules in alleviating gastrointestinal dysfunction in sepsis model rats.Methods The cecal ligation and puncture(CLP)method was used to establish a sepsis rat model.Rats were randomly divided into the sham-operation group,the CLP group,the low dose Shenqihuang group,the medium dose Shenqihuang group,the high dose Shenqihuang group and the dexamethasone group.After modeling,animals were treated for three consecutive days.Rats in the low dose Shenqihuang group,the medium dose Shenqihuang group,the high dose Shenqihuang group were administered Shenqihuang Granules by gavage.The dexamethasone group received dexamethasone by gavage,and the remaining groups were given an equal volume of distilled water by gavage.Enzyme-linked immunosorbent assay was used to detect serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),anti-inflammatory factors interleukin-10(IL-10)and arginase-1(Arg-1),and intestinal injury markers diamine oxidase(DAO)and D-lactic acid(D-Lac).Hematoxylin-Eosin(HE)staining was used to observe the histopathological changes in colonic tissue.The gut microbiome is sequenced using metagenomics to analyze the compositional differences in bacterial phyla across groups and identify characteristic microbes.Results Shenqihuang Granules significantly down-regulated serum pro-inflammatory cytokines IL-6 and TNF-α(P<0.01,P<0.05)and elevated anti-inflammatory factors IL-10 and Arg-1(P<0.01,P<0.05).It also improved colon histopathological changes,down-regulated intestinal injury markers DAO and D-Lac(P<0.01),and modulated gut microbiota to restore microbial balance.Conclusion Shenqihuang Granules offer therapeutic benefits for sepsis rats by regulating inflammation,alleviating intestinal injuries,and modulating gut microbiota.
