为了探究黄芩苷对伪狂犬病毒(Pseudorabies virus,PRV)感染大鼠睾丸及血睾屏障的影响,试验将 30 只 SPF级 7 周龄 SD 大鼠随机分为 5 组,分别为对照组(Control 组)、PRV 组、黄芩苷低剂量组(BAI-L 组,40 mg/kg)、黄芩苷中剂量组(BAI-M 组,80 mg/kg)、黄芩苷高剂量组(BAI-H 组,160 mg/kg),每组 6 只.Control 组和 PRV 组大鼠连续14 d 灌胃 PBS 2 mL,BAI-L、BAI-M、BAI-H 组分别按体重灌胃 40,80,160 mg/kg 黄芩苷,每天 2 mL;第 15 天 Control组大鼠滴鼻生理盐水60 μL,其余大鼠滴鼻PRV-XD-F3 悬液60 μL;然后Control 组和PRV 组大鼠灌胃PBS 2 mL,BAI各剂量组按照不同剂量灌胃黄芩苷溶液2 mL,连续3 d,在第17 天灌胃后全部处死,采集血液及睾丸.H.E.染色观察睾丸组织损伤情况,用试剂盒检测睾丸组织中炎症因子、抗氧化指标和睾丸活性标志酶并检测血清中睾酮浓度,采用实时荧光定量 PCR 检测睾丸组织病毒载量,采用免疫组织化学方法检测血睾屏障相关蛋白.结果表明:与 Control 组相比,PRV 组睾丸间质细胞减少,间隙明显增大并出现空腔;BAI 各剂量组的睾丸间质间隙比 Control 组大,但间质间隙比 PRV 组小,且 BAI-H 组改善效果最好.与 Control 组相比,PRV 组白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)质量浓度,丙二醛(MDA)浓度,活性氧(ROS)浓度均显著升高(P<0.05),白细胞介素-10(IL-10)质量浓度,超氧化物歧化酶(SOD)活性,睾丸活性标志酶酸性磷酸酶(ACP)、碱性磷酸酶(ALP)、葡萄糖-6-磷酸脱氢酶(G-6-PD)、乳酸脱氢同工酶 X(LDH-X)质量浓度及睾酮浓度均显著降低(P<0.05).与 PRV 组相比,BAI-L 组和 BAI-M 组 IL-1β、IL-6 质量浓度显著降低(P<0.05),IL-10 质量浓度显著升高(P<0.05);BAI-H组 IL-1β、IL-6、TNF-α 质量浓度显著降低(P<0.05),IL-10 质量浓度显著升高(P<0.05);BAI-L 组、BAI-M 组和BAI-H 组 MDA、ROS 浓度显著降低(P<0.05),SOD 活性显著升高(P<0.05);BAI-L 组、BAI-M 组和 BAI-H 组 ACP、G-6-PD、LDH-X、睾酮质量浓度显著升高(P<0.05),PRV 拷贝数显著降低(P<0.05),BAI-M 组 ALP 质量浓度显著升高(P<0.05);黄芩苷上调了 PRV 感染后血睾屏障中 ZO-1、Claudin-11、Connexin-43和 N-Cadherin 蛋白的表达.说明黄芩苷可能通过抑制 PRV 复制、降低炎症反应和氧化应激、修复血睾屏障损伤有效缓解 PRV 引起的病毒性睾丸损伤.

In order to investigate the effects of baicalin on testicular tissue and the blood-testis barrier(BTB)in rats infected with Pseudorabies virus(PRV),30 SPF-grade 7-week-old SD rats were randomly divided into five groups(n=6 per group),which were the Control group,PRV group,low-dose baicalin group(BAI-L,40 mg/kg),medium-dose baicalin group(BAI-M,80 mg/kg),and high-dose baicalin group(BAI-H,160 mg/kg).Rats in the Control and PRV groups were orally administered 2 mL of PBS daily for 14 consecutive days,while the BAI-L,BAI-M,and BAI-H groups received 2 mL of baicalin solution at doses of 40,80,and 160 mg/kg,respectively.On day 15,all groups except the Control group were intranasally inoculated with 60 μL of PRV-XD-F3 suspension,whereas the Control group received an equal volume of normal saline.Subsequently,the Control and PRV groups continued to receive 2 mL of PBS daily for three days,while the BAI groups were administered their respective doses of baicalin solution.On day 17,all rats were euthanized,and blood and testicular samples were collected.Testicular tissue damage was assessed by H.E.staining.Inflammatory cytokines,antioxidant indexes,and testicular activity-related enzymes in testicular tissue were measured using assay kits,and serum testosterone levels were quantified.Viral load in testicular tissue was determined by quantitative real-time PCR.Immunohistochemistry was performed to evaluate the expression of BTB-associated proteins.The results showed that compared with the Control group,the PRV group exhibited a reduction in leydig cells,significantly enlarged intercellular spaces,and cavity formation.All BAI groups showed larger interstitial spaces than the Control group,but smaller spaces than the PRV group,with the BAI-H group demonstrating the most pronounced improvement.Compared with the Control group,the PRV group displayed significantly increased levels of interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),malondialdehyde(MDA),and reactive oxygen species(ROS)concentration(P<0.05),along with significantly decreased levels of interleukin-10(IL-10),superoxide dismutase(SOD)activity,testicular activity-related enzymes acid phosphatase(ACP),alkaline phosphatase(ALP),glucose-6-phosphate dehydrogenase(G-6-PD),lactate dehydrogenase isoenzyme X(LDH-X),and testosterone concentration(P<0.05).Compared with the PRV group,the mass concentrations of IL-1β and IL-6 in BAI-L and BAI-M groups significantly decreased(P<0.05)and that of IL-10 significantly increased(P<0.05).The mass concentrations of IL-1β,IL-6,and TNF-α in BAI-H group significantly decreased(P<0.05)and that of IL-10 significantly increased(P<0.05).All baicalin-treated groups(BAI-L,BAI-M,and BAI-H)demonstrated significantly reduced MDA concentration and ROS concentration(P<0.05),and SOD activity significantly increased(P<0.05).Furthermore,these levels of ACP,G-6-PD,LDH-X,and testosterone in baicalin-treated groups(BAI-L,BAI-M,and BAI-H)significantly increased(P<0.05),along with a significant reduction in PRV copy number(P<0.05).The BAI-M group also exhibited a significant increase in ALP levels(P<0.05).Baicalin increased the expression of ZO-1,Claudin-11,Connexin-43,and N-Cadherin proteins in the blood-testis barrier following PRV infection.The results suggested that baicalin might effectively alleviate PRV-induced viral testicular damage by inhibiting viral replication,reducing inflammatory responses and oxidative stress,and repairing BTB integrity.
