Research progress on the application of nanobodies in the diagnosis,treatment and vaccine development of swine diseases

猪病不仅严重制约全球养猪业的健康发展,更对经济发展、食品安全及公共卫生构成多重威胁.然而,当前的传统防控手段,包括诊断技术、治疗方法和疫苗预防,在灵敏度、成本、广谱性、组织穿透性及应对高变异病原等方面仍存在明显局限,难以满足高效防控的需求.纳米抗体作为源自骆驼科动物重链抗体的新型基因工程抗体,具备分子量小、稳定性高、亲和力强、易于大规模制备及功能化改造等独特优势,为猪病防控领域带来突破性解决方案.本文系统阐述纳米抗体的结构特性,重点总结其在猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS)、非洲猪瘟(african swine fever,ASF)、猪流行性腹泻(porcine epidemic diarrhea,PED)、猪瘟(classical swine fever,CSF)和猪圆环病毒病(porcine circovirus disease,PCVD)等重要猪病诊断、治疗及疫苗研发中的应用研究进展,以期为猪病的高效防控提供新思路与理论依据.

Porcine diseases severely restrict the development of the global swine industry and pose multiple threats to economic development,food safety,and public health.However,current traditional prevention and control technologies,including diagnostic techniques,therapeutic approaches,and vaccine prevention,still have significant limitations in terms of sensitivity,cost,broad-spectrum capability,tissue penetration,and the response to highly variable pathogens,which could not meet the demands of prevention and control.Nanobodies(Nbs),a novel class of genetically engineered antibodies derived from heavy-chain antibodies of camelids,present a promising solution for porcine disease control due to their unique advantages,including small molecular size,high stability,strong binding affinity,ease of large-scale production,and engineering modification.This review systematically discussed the structure and characteristics of nanobodies and comprehensively reviewed their application progress in diagnostic technologies,therapeutic strategies,and vaccine development for major porcine diseases,such as porcine reproductive and respiratory syndrome(PRRS),African swine fever(ASF),porcine epidemic diarrhea(PED),classical swine fever(CSF),and porcine circovirus disease(PCVD),aiming to provide new insights and theoretical foundations for the efficient prevention and control of porcine diseases.
