山奈酚通过激活Wnt/β-catenin通路抑制神经干细胞凋亡改善脑梗死预后的研究OA
Kaempferol inhibits neural stem cell apoptosis by activating the Wnt/β-catenin pathway and improve the prognosis of cerebral infarction
目的 探讨山奈酚通过激活Wnt/β-catenin通路抑制神经干细胞(NSCs)凋亡改善脑梗死预后的影响.方法 在体内实验中,通过给予大脑中动脉闭塞(MCAO)小鼠模型山奈酚治疗,测定山奈酚对梗死体积和神经功能的影响,比较各组Western blot测定Wnt/β-catenin通路相关蛋白的变化了解其作用机制.建立NSCs氧糖剥夺(OGD)/再氧化(R)模型,通过细胞计数试剂盒-8、Transwell实验、流式细胞术分析评估山奈酚对神经干细胞细胞活力、迁移和凋亡的影响.结果 山奈酚改善脑梗死小鼠神经功能缺损,减少梗死体积,促进神经再生(P<0.05).其次,山奈酚还能促进OGD/R处理的NSCs的细胞活力和迁移,抑制细胞凋亡(P<0.05).此外,山奈酚提高Wnt/β-catenin通路活性,而阻断Wnt/β-catenin通路可以引起山奈酚介导的NSCs的生物学功能丧失(P<0.05).结论 山奈酚通过促进NSCs活力、迁移和抑制细胞凋亡抑制脑梗死,山奈酚通过激活Wnt/β-catenin通路改善脑梗死后神经功能损伤,有望成为缺血性脑梗死治疗的新药物.
Objective To investigate the effect of kaempferol on inhibiting apoptosis of neural stem cells by activating the Wnt/β-catenin pathway to improve the prognosis of cerebral infarction.Methods In vivo,kaempferol was administered to middle cerebral artery occlusion(MCAO)mouse model to determine the effects of kaempferol on infarct volume and nerve function,and the changes of Wnt/β-catenin pathway related proteins in each group were compared by western blot analysis to understand the mechanism of action.OGD/reoxidation(R)models of neural stem cells(NSCs)were established,and the effects of kaempferol on cell viability,migration and apoptosis were evaluated by cell counting kit 8,Transwell assay,flow cytometry and TUNEL analysis.Results Kaempferol improved nerve function defect,reduced infarct volume and promoted nerve regeneration in mice with cerebral infarction.Secondly,kaempferol can also promote the cell viability and migration of OGD/R treated NSCs and inhibit cell apoptosis.In addition,kaempferol enhances the activity of the Wnt/β-catenin pathway,and blocking the Wnt/β-catenin pathway can cause the loss of biological function of kaempferol-mediated NSCs.Conclusion Kaempferol can inhibit cerebral infarction by promoting NSCs activity,migration and inhibiting apoptosis.Kaempferol can improve nerve function injury after cerebral infarction by activating Wnt/β-catenin pathway,which is expected to be a new drug for the treatment of ischemic cerebral infarction.
