非洲鸵鸟胸腺内生长抑素的分布特点OA
Distribution characteristics of somatostatin in the thymus of African ostriches
为探究非洲鸵鸟胸腺内生长抑素(somatostatin,SST)的分布特点,试验选择 1 日龄、45 日龄、90 日龄健康育雏期非洲鸵鸟和 334 日龄健康成年非洲鸵鸟各 6 羽,麻醉后摘取胸腺组织并制作石蜡切片,采用免疫组织化学技术检测胸腺内的 STT 阳性细胞,统计单位面积 SST 阳性细胞数量,并观察 90 日龄非洲鸵鸟胸腺内 STT 阳性细胞的分布与形态学特征.结果表明:不同日龄非洲鸵鸟胸腺内均存在 SST 阳性细胞.随着日龄的增加,胸腺内单位面积 SST 阳性细胞数量呈升高趋势.与 1 日龄相比,45 日龄、90 日龄、334 日龄非洲鸵鸟胸腺内单位面积 SST 阳性细胞数量显著升高(P<0.05);90 日龄和 334 日龄非洲鸵鸟胸腺内单位面积 SST 阳性细胞数量较45 日龄虽有小幅升高趋势,但差异不显著(P>0.05).90 日龄非洲鸵鸟胸腺内 SST 阳性细胞呈散在分布,主要集中在胸腺皮质和髓质交界区域,多数 SST 阳性细胞呈圆形或锥状,少数 SST 阳性细胞呈梭形或梨形并伴有细长胞质突起.说明非洲鸵鸟胸腺内 SST 表达在育雏期早期阶段迅速增加,随后趋于稳定;SST 阳性细胞主要分布于胸腺皮质和髓质交界区域.
In order to explore the distribution of somatostatin(SST)in the thymus of African ostriches,6 healthy African ostriches aged 1-day-old,45-day-old,and 90-day-old in rearing period,as well as 6 adult African ostriches aged 334-day-old,were selected.After anesthesia,thymus tissues were collected and processed into paraffin sections.Immunohistochemistry was used to detect SST-positive cells within the thymus.The number of SST-positive cells per unit area was counted,and the distribution patterns and morphological features of these cells were observed specifically in the 90-day-old ostriches.The results showed that SST-positive cells were present in the thymus of African ostriches at all examined ages.With the increase of age,the number of SST-positive cells per unit area in the thymus showed an upward trend.Compared with the 1-day-old group,the number of SST-positive cells per unit area was significantly increased in the 45-day-old,90-day-old,and 334-day-old groups(P<0.05).Although there was a slight increase in the number of SST-positive cells per unit area in the thymus of African ostriches at 90-day-old and 334-day-old of age compared with 45-day-old,the differences were not significant(P>0.05).In 90-day-old ostriches,SST-positive cells were scattered throughout the thymus and primarily concentrated in border area between the thymic cortex and medulla,most SST-postive cells appeared round or conical in shape,while a few SST-postive cells were spindle-shaped or pear-shaped,accompanied by slender cytoplasmic projections.These findings indicated that SST expression in the thymus of African ostriches increased rapidly during the early brooding stage and subsequently stabilizes and then stabilized.SST-positive cells were mainly distributed in the corticomedullary junction region of the thymus.
