混合盐碱胁迫对羊草种子萌发的影响OA
Effects of mixed saline-alkali stress on seed germination of Leymus chinensis
为了探究混合盐碱胁迫对羊草种子萌发的影响,试验以中性盐(NaCl、Na2SO4)与碱性盐(NaHCO3、Na2CO3)按照摩尔比 1∶9∶9∶1[轻度(S),pH 值=9.07]、1∶1∶1∶1[中度(M),pH 值=9.70]、9∶1∶1∶9[(重度(L),pH值=10.24]模拟盐碱胁迫环境,每个处理设置 5 个浓度梯度(30,60,90,120,150 mmol/L),以纯化水为空白对照(CK,pH 值=6.90),对 9 号羊草种子进行盐碱胁迫,测定羊草种子的萌发指标(发芽率、发芽势、发芽指数、活力指数)、幼苗形态指标(胚根长、胚芽长)及盐害率,计算羊草种子萌发期适宜盐浓度、半致死浓度和极限浓度,通过盐浓度、pH 值及其交互作用对羊草种子萌发进行双因素方差分析.结果表明:低盐浓度(30 mmol/L)、轻度(pH 值=9.07)混合盐碱胁迫对羊草种子的萌发无显著影响,随着盐浓度和 pH 值的升高,羊草种子萌发抑制作用逐渐增强.当盐浓度达 150 mmol/L,重度(pH 值=10.24)混合盐碱胁迫时,羊草种子几乎不能萌发.当 pH 值为 10.24 时,羊草种子适宜盐浓度仅为 37.95 mmol/L.盐浓度、pH 值及盐浓度与 pH 值交互作用均会显著抑制羊草种子萌发,其中 pH 值是决定性的影响因素,其次是盐浓度.说明在混合盐碱环境中,酸碱性因素在抑制羊草种子萌发方面起主导作用.
In order to investigate the effects of mixed saline-alkali stress on seed germination of Leymus chinensis(L.chinensis),the experiment simulated saline-alkali stress environments using neutral salts(NaCl,Na2 SO4)and alkaline salts(NaHCO3,Na2 CO3)were mixed in molar ratios of 1∶9∶9∶1(mild[S],pH value=9.07),1∶1∶1∶1(moderate[M],pH value=9.70),and 9∶1∶1∶9(severe[L],pH value=10.24).Each treatment group included five concentration gradients(30,60,90,120,150 mmol/L),with purified water as the control(CK,pH value=6.90).The seeds of L.chinensis No.9 were subjected to saline-alkali stress,and germination indicators(germination rate,germination potential,germination index,vigor index),morphological indicators of seedlings(radicle length,plumule length)and salt damage rate were measured,and a two-way analysis of variance was conducted on the seed germination of L.chinensis to examine the effects of salt concentration,pH value,and their interaction.The suitable mixed salt concentration range,half-lethal concentration,and limit concentration of L.chinensis seeds germination were calculated.The results showed that low salt concentration(30 mmol/L)and mild(pH value=9.07)mixed saline-alkali stress had no significant effect on the germination of L.chinensis seeds.However,as the salt concentration and pH increased,the inhibitory effect on the germination of L.chinensis seeds gradually intensified.When the salt concentration reached 150 mmol/L and under severe(pH=10.24)mixed saline-alkali stress,L.chinensis seeds could hardly germinate.When the pH value was 10.24,the suitable salt concentration of L.chinensis seed germination was only 37.95 mmol/L.It was found that salt concentration,pH value and their interaction all significantly inhibited the germination of L.chinensis seeds,among which pH value was the decisive influencing factor,followed by salt concentration.It indicated that in the mixed saline-alkali environment,the alkaline factor(pH value)played a dominant role in inhibiting the seed germination of L.chinensis.
