七氟烷麻醉对青光眼大鼠视网膜神经节细胞凋亡和视网膜影响的实验研究OA
Experimental Study on the Effects of Sevoflurane Anaesthesia on Retinal Ganglion Cell Apoptosis and Retinal Function in Glaucoma Rats
目的:探究青光眼大鼠行七氟烷麻醉后对视网膜神经节细胞凋亡及视网膜的影响.方法:选取40只大鼠随机分为假手术组12只,剩余28只建立青光眼大鼠模型,其中失败4只,剩余24只随机分为模型组12只,七氟烷组12只.对比各组(不同时间)眼压变化、病理学变化、视网膜厚度、神经节数量、氧化应激指标、视网膜形态学细胞密度、线粒体受损情况、视网膜神经节细胞凋亡数量及内质网应激途径.结果:与模型组相比,七氟烷组P/O、外丛状层、Cytc/a、神经节细胞层、H2O2、神经节数量、内丛状层、视网膜厚度、外核层升高,眼压MDA、凋亡细胞数、SOD、GRP78、ROS、CHOP水平下降,差异具有统计学意义(P<0.05).结论:七氟烷麻醉可降低青光眼大鼠视网膜神经节细胞凋亡,改善氧化应激、线粒体受损、内质网应激情况,提高神经节数量,增加视网膜厚度、视网膜形态学细胞密度,进而对视网膜神经纤维层发挥保护作用.
Objective:To investigate the effects of sevoflurane anesthesia on the apoptosis of retinal gan-glion cells and the retina in glaucoma rats.Methods:Twelve rats were randomly seperated from forty to make a sham operation group.The other 28 rats were used to establish glaucoma rat models,among which 4 failed.The remaining 24 rats were randomly divided into a model group of 12 rats and a sevoflurane group of 12 rats.The intraocular pressure changes,pathological changes,retinal thickness,ganglion number,oxidative stress indicators,retinal morphological cell density,mitochondrial damage,the number of retinal ganglion cell apop-tosis and endoplasmic reticulum stress pathways in each group(at different times)were compared.Results:After modeling,the intraocular pressure in the three groups of rats showed a gradually increasing trend over time.Compared with the sham operation group,the intraocular pressure in the model group and the sevoflu-rane group(1 day,2 days,4 days,and 7 days after modeling)increased,and the difference was statistical-ly significant(P<0.05).Compared with the sham operation group,in the sevoflurane group and the model group,the levels of the outer nuclear layer,Cytc/a,inner plexiform layer,P/O,retinal thickness,H2O2,outer plexiform layer,ganglion number,and ganglion cell layer decreased,while the levels of ROS,apoptotic cells,SOD,GRP78,MDA,and CHOP increased.Compared with the model group,the levels of P/O,outer plexiform layer,Cytc/a,ganglion cell layer,H2O2,ganglion number,inner plexiform layer,retinal thick-ness,and outer nuclear layer in the sevoflurane group increased,while the levels of MDA,apoptotic cells,SOD,GRP78,ROS,and CHOP decreased.The differences were statistically significant(P<0.05).Con-clusion:Sevoflurane anesthesia can reduce the apoptosis of retinal ganglion cells in glaucoma rats,improve oxidative stress,mitochondrial damage and endoplasmic reticulum stress,increase the number of ganglia,en-hance retinal thickness and the density of retinal morphological cells,and thereby exert a protective effect on the retinal nerve fiber layer.
