资癸益冲方通过VEGF/Notch1信号调节卵巢血管生成和自噬改善大鼠卵巢储备功能下降OA
Zigui Yichong Formula Improves Diminished Ovarian Reserve in Rats by Regulating Ovarian Angiogenesis and Autophagy Through VEGF/Notch1 Signaling
目的:探究资癸益冲方对DOR大鼠的卵巢血管生成的作用机制,并分析与VEGF/Notch1信号的关系.方法:选取SPF级雌性SD大鼠40只作为实验动物,经阴道脱落细胞涂片确定33只动情周期规律的大鼠,并将其随机分为空白对照组、模型组、资癸益冲方组、资癸益冲方+Axitinib组各8只.对比各组卵巢湿重、卵巢指数、形态学观察、性激素、卵泡数目、卵巢血管生成情况、自噬及相关蛋白表达及VEGF/Notch1信号表达.结果:与空白对照组相比,模型组、资癸益冲方+Axitinib组初级卵泡、AMH、窦状卵泡、VEGF、卵巢湿重、E2、卵巢指数、原始卵泡、CD31下降,Lc3 Ⅱ、FSH、闭锁卵泡、Beclin1、LH、Notch 1、自噬溶酶体数量水平升高;与模型组、资癸益冲方+Axitinib组相比,资癸益冲方组CD31、初级卵泡、VEGF、卵巢指数、E2、原始卵泡、卵巢湿重、窦状卵泡、AMH升高,Beclin1、LH、Lc3 Ⅱ、闭锁卵泡、Notch 1、自噬溶酶体数量、FSH水平下降,差异具有统计学意义(P<0.05).结论:资癸益冲方可通过调节VEGF/Notch1信号通路调节性激素、促进卵巢血管生成、降低自噬等多途径改善DOR大鼠卵巢储备功能.
Objective:To investigate the mechanism of Zigui Yichong Formula on ovarian angiogenesis in rats with diminished ovarian reserve,and analyze its relationship with VEGF/Notch1 signaling.Methods:Forty SPF-grade female SD rats were selected as experimental animals.33 rats with regular estrous cycles were determined by vaginal exfoliated cell smears and randomly divided into blank control group,model group,Zigui Yichong formula group and Zigui Yichong formula+Axitinib group,with 8 rats in each group.The ovarian wet weight,ovarian index,morphological observation,sex hormones,number of follicles,Ovarian angiogenesis,autophagy and related protein expression,as well as VEGF/Notch1 signal expression of each group were com-pared.Results:Compared with the blank control group,primary follicles,AMH,sinus follicles,VEGF,o-varian wet weight,E2,ovarian index,primordial follicles,and CD31 were decreased,and Lc3 Ⅱ,FSH,atretic follicles,Beclin1,LH,Notch1,and autophagy lysosome number levels were increased in the model and ZiqiYiFong Formula+Axitinib groups.Compared with the model group and Zigui Yichong Formul+Ax-itinib group,in the Zigui Yichong Formula group,CD31,primary follicle,VEGF,ovarian index,E2,primor-dial follicle,ovarian wet weight,sinusoidal follicle,and AMH levels were increased,and the levels of Bec-lin1,LH,Lc3 Ⅱ,atretic follicle,Notch 1,autophagy lysosome number,and FSH levels were decreased,there was statistically significant differences(P<0.05).Conclusion:Zigui Yichong Formula can improve ovarian reserve in DOR rats through multiple pathways,including regulating sex hormones,promoting ovarian angio-genesis,and reducing autophagy,by modulating the VEGF/Notch1 signaling pathway.
