枳实水提物的降脂活性OA
Hypolipidemic Activity of the Water Extract of Aurantii Fructus Immaturus
为探讨枳实水提物对非酒精性脂肪肝(NAFLD)细胞模型的降脂作用及氧化应激调节机制,本研究利用水回流提取法获得枳实水提物,采用CCK-8法测定枳实水提物对HepG2细胞的毒性;降脂试验则以游离脂肪酸(FFA)诱导构建NAFLD模型,设置空白对照组、模型组、枳实水提物干预组(25、50、100 µg/mL)及水飞蓟素阳性药物组(25 µg/mL).通过试剂盒检测细胞内甘油三酯(TG)和总胆固醇(TC)含量;利用油红O染色观察脂滴堆积,并测定超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽(GSH)、丙二醛(MDA)以及活性氧(ROS)水平.结果显示,25~400 µg/mL枳实水提物对HepG2细胞无显著毒性(细胞存活率>94%);与模型组相比,50、100 µg/mL枳实水提物分别降低细胞内TG含量28.6%、41.2%,TC含量分别减少25.3%、38.7%,减少脂滴堆积且呈浓度依赖性,其中100 µg/mL组降脂效果与水飞蓟素相当;同时,50、100 µg/mL枳实水提物可分别显著提升SOD活性21.5%、32.8%,提高CAT活性18.7%、29.4%及升高GSH含量23.1%、35.6%,降低MDA含量26.8%、40.3%和减少ROS含量30.2%、42.5%.综上所述,枳实水提物通过调节氧化应激与脂质代谢发挥降脂作用,为NAFLD防治提供实验依据.
To investigate the lipid-lowering effects and oxidative stress modulation mechanisms of Aurantii Fructus Immaturus water extract(AFIWE)on a cellular model of non-alcoholic fatty liver disease(NAFLD),this study obtained AFIWE through water reflux extraction.The cytotoxicity of AFIWE on HepG2 cells was assessed using the CCK-8 assay.For the lipid-lowering experiment,an NAFLD model was established by free fatty acid(FFA)induction,with experimental groups including a blank control group,model group,AFIWE intervention groups(25,50 and 100 µg/mL),and a positive control group(25 µg/mL silymarin).Intracellular triglyceride(TG)and total cholesterol(TC)levels were measured using commercial assay kits.Lipid droplet accumulation was observed via Oil Red O staining,and the activities of superoxide dismutase(SOD),catalase(CAT),glutathione(GSH),as well as the levels of malondialdehyde(MDA)and reactive oxygen species(ROS),were determined.Results demonstrated that AFIWE at concentrations ranging from 25 to 400 µg/mL exhibited no significant cytotoxicity(cell viability>94%).Compared with the model group,AFIWE at 50 and 100 µg/mL reduced intracellular TG content by 28.6%and 41.2%,and TC content by 25.3%and 38.7%,respectively.Lipid droplet accumulation was also reduced in a concentration-dependent manner,with the 100 µg/mL group showing lipid-lowering effects comparable to those of silymarin(p>0.05).Furthermore,AFIWE at 50 and 100 µg/mL significantly enhanced SOD activity by 21.5%and 32.8%,increased CAT activity by 18.7%and 29.4%,elevated GSH content by 23.1%and 35.6%,reduced MDA levels by 26.8%and 40.3%,and decreased ROS levels by 30.2%and 42.5%,respectively.In conclusion,AFIWE exerted lipid-lowering effects by regulating oxidative stress and lipid metabolism,providing experimental evidence for the prevention and treatment of NAFLD.
