目的:探讨五味子乙素(Sch B)对咳嗽变异型哮喘(CVA)大鼠治疗及CC趋化因子配体2(CCL2)/CC类趋化因子受体2(CCR2)信号轴的影响.方法:构建CVA大鼠模型,将大鼠分为对照组(Control 组)、模型组(CVA 组)、Sch B 低、中、高剂量组(Sch B-L、Sch B-M、Sch B-H 组)、Sch B 高剂量+CCL2重组蛋白(Sch B-H+CCL2组).检测大鼠咳嗽次数、1s钟用力呼气容积(FEV1)、FEV1/用力肺活量(FVC)(FEV1/FVC)及肺泡灌洗液中白介素-4(IL-4)、白介素-5(IL-5)、干扰素-γ(IFN-γ)、白三烯C4(LTC4)水平和嗜酸粒细胞计数(EOS);观察肺组织病理变化;检测肺组织骨桥蛋白(OPN)、CCL2、CCR2表达水平.结果:CVA组大鼠肺组织损伤严重,咳嗽次数增多、IL-4、IL-5、EOS、LTC4、OPN、CCL2、CCR2 表达升高,FEV1、FEV1/FVC、PEF、IFN-γ 水平降低(P<0.05);Sch B 可减轻大鼠肺组织损伤,减少咳嗽次数,降低 IL-4、IL-5、EOS、LTC4、OPN、CCL2、CCR2 表达,升高 FEV1、FEV1/FVC、PEF、IFN-γ水平(P<0.05);CCL2重组蛋白可部分逆转Sch B对CVA大鼠治疗作用.结论:Sch B可能通过抑制CCL2-CCR2信号轴治疗CVA大鼠.

Objective:To investigate the effects of schisandrin(SchB)on cough variant asthma(CVA)rats and CC chemokine ligand 2(CCL2)/CC-like chemokine receptor 2(CCR2)signaling axis.Methods:The CVA rat model was established,and the rats were divided into Control group,model group,Schisandra B low,medium and high dose groups(SchB-L,SchB-M and SchB-H groups)and schisandra B high dose+CCL2 recombinant protein(SchB-H+CCL2 group).The cough frequency,forced expiratory volume(FEV1),FEV1/forced vital capacity(FVC)(FEV1/FVC)ratio,the levels of interleukin-4(IL-4),in-terleukin-5(IL-5),interferon-γ(IFN-γ)and leutriene C4(LTC4)as well as eosinophilic count(EOS)in alveolar lavage fluid were detected.The pathological changes of lung tissue were observed.The expression levels of osteopontin(OPN),CCL2 and CCR2 in lung tissue were detected.Results:In CVA group,lung tissue injury was serious,cough frequency was increased,expressions of IL-4,IL-5,EOS,LTC4,OPN,CCL2,CCR2 were increased,and levels of FEV1,FEV1/FVC,PEF,IFN-γ were decreased(P<0.05);SchB could alleviate lung tissue injury,reduce the number of cough,decrease the expression of IL-4,IL-5,EOS,LTC4,OPN,CCL2,CCR2,and increase the levels of FEV1,FEV1/FVC,PEF,IFN-γ(P<0.05);CCL2 recombinant protein could partially reverse the therapeutic effect of Sch B on CVA rats(P<0.05).Conclusion:Schisandrin B may exert therapeutic effects on CVA rats by inhibiting the CCL2-CCR2 signaling axis.
