目的:通过建立大鼠DMCP模型,探究丹酚酸B(salB)对其治疗作用以及对PTEN/Nrf2信号通路的影响.方法:58只雄性SD大鼠被随机分配到对照组(CON,n=8):DMCP模型组(DMCP,n=50),采用STZ诱导DM大鼠并结合上颌结扎诱导DMCP模型.将成功建模的43只大鼠随机分成:DMCP组(n=11)、低、中、高剂量salB治疗组(salB-L、salB-M、salB-H,每组分别8只)、阳性对照组(二甲双胍,MET,n=8).各组分别每日灌胃给予相应药物或等体积生理盐水,持续6周.采用活体荧光成像技术评估体内ROS水平;在Micro-CT图像中,CEJ与ABC之间的直线距离来量化牙槽骨缺损程度;使用H&E和Masson染色进行组织学分析;使用ELISA试剂盒测定结扎部位周围牙龈组织中促炎细胞因子(TNF-α、IL-1β、IL-6、IL-17)水平;采用免疫荧光染色法检测巨噬细胞极化对牙周炎发展的影响;RT-PCR和 Western blot技术测定PTEN和Nrf2的表达情况.结果:与CON组相比,DMCP组大鼠CEJ-ABC间距显著增加,差异具有统计学意义(P<0.05);组织学分析结果表明DMCP组大鼠牙槽骨损伤严重并伴有炎症反应;且ROS以及促炎因子(TNF-α、IL-1β、IL-6、IL-17)水平显著升高,差异具有统计学意义(P<0.05);RT-PCR和 Western blot结果表明DMCP组大鼠PTEN表达降低、Nrf2的表达升高,差异具有统计学意义(P<0.05).与DMCP组相比,中、高剂量salB组大鼠CEJ-ABC间距显著减少,恢复牙槽骨损伤,ROS以及促炎因子水平显著降低,差异具有统计学意义(P<0.05);salB治疗后PTEN表达上调、Nrf2的表达更加上调,差异具有统计学意义(P<0.05).结论:salB能缓解DMCP对牙槽骨的破坏作用,可降低ROS水平、调节巨噬细胞极化和炎性因子水平,并且可能与其对PTEN/Nrf2通路的调节有关.

Objective:To investigate the therapeutic effect of salvianolic Acid B(salB)and its impact on the PTEN/Nrf2 signaling pathway in a rat model of DMCP.Methods:A total of 38 male SD rats were ran-domly assigned to control group(CON,n=8)and DMCP model group(DMCP,n=30).The DMCP model was established by inducing DM with streptozotocin and combining it with maxillary ligation to induce CP.The 25 successfully modeled rats were then randomly divided into:DMCP group(n=9),salB treatment group(n=8),and positive control group(metformin,MET,n=8).Each group was administered the corresponding drug or an equal volume of normal saline via gastric intubation daily for 6 weeks.In vivo ROS levels were as-sessed using live fluorescence imaging.Alveolar bone defect severity was quantified on micro-CT images by measuring the linear distance between the CEJ and the ABC.Histological analysis was performed using H&E and Masson staining.Levels of pro-inflammatory cytokines(TNF-α,IL-1β,IL-6,IL-17)in the gingival tissue surrounding the ligature site were measured using ELISA kits.The impact of macrophage polarization on periodontitis development was detected using immunofluorescence staining.Expression levels of PTEN and Nrf2 were determined by RT-PCR and Western blot.Results:Compared to the CON group,the DMCP group showed a statistically significant increase in the CEJ-ABC distance(P<0.05).Histological analysis revealed severe alveolar bone damage accompanied by an inflammatory response in the DMCP group.ROS and pro-in-flammatory cytokine(TNF-α,IL-1β,IL-6,IL-17)levels were significantly elevated in the DMCP group(P<0.05).RT-PCR and Western blot results indicated a statistically significant decrease in PTEN expres-sion and an increase in Nrf2 expression in the DMCP group(P<0.05).Compared to the DMCP group,the salB-M and salB-H groups exhibited a statistically significant reduction in the CEJ-ABC distance,restoration of alveolar bone damage,and significantly lower levels of ROS and pro-inflammatory cytokines(P<0.05).After salB treatment,PTEN expression was upregulated and Nrf2 expression was further upregulated,with sta-tistical significance(P<0.05).Conclusion:SalB alleviated the destructive effects of DMCP on alveolar bone,reduced ROS levels,modulated macrophage polarization and inflammatory cytokine levels,which might be related to its regulation of PTEN/Nrf2 pathway.
