预知子提取物的不同分离部位对脂多糖诱导BV2细胞介导神经炎症作用及其机制的研究OA
Mechanism of Action of Different Isolated Parts of Akebiae Fructus Extract on LPS-Induced Neuro-Inflammation in BV2 Cells
目的 探讨预知子提取物不同分离部位对脂多糖(LPS)诱导的神经炎症反应的影响及其潜在机制.方法 通过乙醇回流法提取预知子提取物(JE),并利用系统溶剂萃取法分离出正己烷层(JE1)、二氯甲烷层(JE2)、乙酸乙酯层(JE3)和正丁醇层(JE4);利用LPS诱导BV2细胞构建炎症模型;采用四甲基偶氮唑蓝(MTT)法评估细胞活力;通过Griess反应测定细胞内NO浓度;使用ELISA法检测细胞内PGE2、IL-6、TNF-α水平;运用Western Blot法检测COX-2、iNOS蛋白和MAPK、NF-κB信号通路相关蛋白表达水平.结果 JE1和JE2显著降低了LPS诱导的BV2细胞中NO、PGE2、IL-6、TNF-α水平及COX-2和iNOS蛋白表达,并且JE1和JE2显著抑制了P38、ERK、JNK和NF-κB蛋白表达.结论 预知子提取物的JE1和JE2部位能够改善LPS诱导的BV2细胞神经炎症反应,其作用机制或许与抑制NF-κB和MAPK信号通路相关.
Objective To investigate the anti-neuroinflammatory effects of different fractions of the Akebiae Fructus extract on lipopoly-saccharide(LPS)-induced BV2 cells and to explore the underlying mechanisms.Methods The Akebiae Fructus extract(JE)was ob-tained by ethanol reflux extraction,and the systematic solvent extraction method was used to separate the n-hexane fraction(JE1),di-chloromethane fraction(JE2),ethyl acetate fraction(JE3),and n-butanol fraction(JE4).An inflammatory model was established using LPS-induced BV2 cells;MTT assay was used to observe the cell viability.The content of NO in cell supernatant was measured by Griess reagent.The levels of PGE2,IL-6 and TNF-α were detected by ELISA kits.Detection of COX-2、iNOS protein and MAPK、NF-κB sig-naling pathway related protein expression levels using Western Blot method.Results Both JE1 and JE2 significantly reduced the levels of NO,PGE2,IL-6,TNF-α and the protein expressions of COX-2 and iNOS in LPS-induced BV2 cells,and also significantly decreased the protein expressions of P38,ERK,JNK,and NF-κB.Conclusion The JE1 and JE2 fractions of the Akebiae Fructus extract can effec-tively prevent LPS-induced neuroinflammatory responses occur in BV2 cells,and the underlying mechanism is associated with the inhibi-tion of the inflammatory NF-κB and MAPK signaling pathways.
