前处理和胚切割方式对小黑麦成熟胚愈伤组织培养的影响OA
Effects of pretreatment and embryo-cutting method on callus culture of mature triticale embryos
高效的组织培养体系是小黑麦(×Triticosecale Wittmack)种质资源繁育和遗传转化研究的重要环节.本研究以小黑麦成熟胚为外植体,研究不同种子消毒处理、切胚方式和浸种方法对愈伤组织诱导及实生芽生长的影响.结果表明,75%乙醇 1 min 结合 20%NaClO 溶液 15 min 的消毒方法出愈率高且染菌率低,是最佳的前处理消毒组合;选用 2,4-D 和氨氯吡啶酸作为浸种液虽然不能明显地提高出愈率,但在浓度为 1 g·L-1 时却会较大幅度降低实生芽率;2/3 胚横切法可显著提高小黑麦愈伤组织的出愈率,与 1 g·L-1 的 2,4-D 组合使用可作为理想的诱导手段.本研究结果为优化小黑麦种质资源提供了技术参考.
Mature triticale embryos were used as explants to investigate the effects of different disinfection,embryo-cutting and seed-soaking methods on callus induction and sprout growth.Various concentrations of ethanol and sodium hypochlorite solution were used for different durations for disinfecting triticale seeds.The callus induction frequency was higher and the rate of bacterial contamination was lower than those in the control when applying 75%ethanol for 1 min and 20%sodium hypochlorite solution for 15 min for disinfecting triticale callus,which was the most effective pretreatment disinfection method.Soaking seeds with 1 g·L-1 2,4-D or picloram solution did not increase the frequency of callus induction but markedly reduced the sprout rate.Transecting two-thirds of the embryos considerably increased the callus induction frequency.Combining this transection with soaking the seeds in 1 g·L-1 2,4-D was found to be an effective preinduction treatment.These results provide technical support for tissue culture and developing triticale germplasm resources.
